摘 要:目的探讨牛膝多糖(ABPS)对糖尿病大鼠肾组织保护作用及其作用机制。方法将Wistar大鼠随机分为5组,正常对照组、模型组、ABPS300、200、100mg·kg-1·d-1组。8周后,苏木素-伊红(HE)染色观察肾组织病理变化,琼脂糖凝胶电泳检测细胞凋亡,反转录-聚合酶链反应(RT-PCR)观察Bcl-2、Fas基因的表达。结果与模型组比较,ABPS组肾组织形态明显改善,肾组织细胞凋亡明显减少,Bcl-2表达上调,Fas表达降低。结论 ABPS对糖尿病大鼠肾组织有明显保护作用,其机制可能与抑制肾组织细胞凋亡及上调Bcl-2,降低Fas基因表达有关。Objective To investigate the protective effect and its mechanism of achyranthes bidentata blume polysaccharides (ABPS) on renal tissue of diabetic rats. Methods Wistar rats were randomized into the normal control, diabetic model group, and three diabetic groups treated with 300, 200 and 100 mg·kg^-1·d^-1 ABPS, respectively. After 8 weeks, pathologic changes of the renal tissue were observed through HE staining, cell apoptosis was detected by agar gel electrophoresis, and the expression of both Bcl-2 and Fas genes was assessed by RT-PCR. Results Compared with diabetic model group, significant improvements with a decrease in the number of apoptotic cells were shown in the morphology of renal tissues in ABPS group. There was an up-regulation of Fas gene expression and a down-regulation of Bcl-2 gene expression. Conclusion ABPS provided significant reno-protective effect in diabetic rats, probably through inhibition of renal cell apoptosis, up-regulation of Bcl-2 expression and down-regulation of Fas gene expressions.
