散血明目片对兔视网膜静脉阻塞模型tPA、PAI-1表达的影响

摘　　要：目的:探讨活血通脉利水明目之散血明目片对兔视网膜静脉阻塞(RVO)模型视网膜组织中组织型纤溶酶原激活物(tPA)、纤溶酶原激活物抑制剂(PAI-1)表达的干预作用。方法:取健康有色家兔36只,每组9只,分为A组:健康空白组,B组:模型组,C组:血栓通组,D组:散血明目片组。采用波长532nm的Nd:YAG激光光凝法建立RVO模型,连续灌胃,实验后第7、14、21d分别处死家兔取材,石蜡包埋切片,用免疫组化法检测视网膜组织中tPA、PAI-1、表达活性。结果:D组与A、B、C三组比较:tPA活性明显高于A、B两组(P<0.05),与C组差异无显著性;PAI-1表达活性明显低于B组(P<0.05),与C组差异无显著性。结论:活血通脉利水明目之散血明目片能够明显提高RVO后视网膜组织tPA表达,降低PAI-1表达,从而抑制血栓形成,改善RVO后视网膜局部微循环,减轻缺血缺氧。Objective： To investigate the effects of SanXue MingMu Tablets （SXMMT） of activating blood, relieving meridians, draining water and brightening eyes on the expression of tPA, PAI-1 of rabbits’ retina vein obstruction （RVO） model. Method： 36 healthy domestic rabbits with color were divided into 4 groups, 9 for each group, i.e. group A： healthy blank group, group B： model group, group C： XueShuangTong group,and group D： SXMMT group. RVO model were made with Nd： YAG laser photocoagulation whose wavelength is 532nm, administered intragastrically continuously .And the rabbits were killed respectively in the 7th, the 14th, and the 21st days after the experiment, cut the rabbits into slices, and bury the slices with paraffin, then test the tPA,PAI-1and activated expression in retinal organization with the method of immunity combination. Result： The comparison between group D and group A, B and C： the tPA action of group D was obviously higher than that of group A and B （P〈0.05）, but no significant difference from that of group C. The action of PAI-1 expression is evidently lower than that of group B （P〈0.05）, but no significant difference from that of group C. Conclusion： SXMMT of activating blood,relieving meridians,draining water and brightening eyes can significantly increase tPA expression of retinal organization after RVO, decrease expression of PAI-1, inhibit the formation of thrombus, improve microcirculation in regional retina after RVO.
