摘 要:目的:观察密蒙花提取物对去势所致干眼症雄兔基础泪液分泌量、泪腺局部炎症因子TGF-β1、IL-1β、TNF-α的影响,探讨密蒙花提取物抗雄激素水平下降所致的干眼症的作用机制。方法:将30只日本大耳雄兔随机分为空白组(A),模型组(B),治疗组(C、D、E),每组6只,对B、C、D、E组行去势术建立动物模型,对C、D、E组分别以密蒙花提取物1倍剂量和2倍剂量以及染料木黄酮连续灌胃1月,对全部实验兔行Schirmer I试验,采用免疫组织化学的方法,对兔泪腺组织中炎症因子TGF-β1、IL-1β、TNF-α进行检测。结果:C、D、E组Schirmer I试验测量值明显高于B组(P(0.01),泪腺导管及腺泡上皮细胞中,IL-1β、TNF-α阳性表达的细胞数均明显低于B组而TGF-β1阳性表达的细胞数均明显高于B组(P(0.01)。以上检测结果C、D组均优于E组(P(0.05)。结论:密蒙花提取物对于雄激素水平所致兔干眼症有良好的实验疗效,其作用机制可能与其中主要成分黄酮类物质拟雄激素效应调节体内性激素水平有关,从而可抑制泪腺局部炎症反应来起到疗效。Objective:To assess the roles of extract of buddleja officinalis in basic tears secretory volume,tear film stability,expression of TGF-β1,IL-1β,TNF-α in lacrimal gland of castrated rabbits with dry eye,and to discuss the therapeutic effects of extract of buddleja officinalis on dry eye caused by gonadal hormones level imbalance.Methods:30 victory rabbits was divided averagely into normal group(A),model group(B),therapy group with single-dose buddleja officinalis extract(C),therapy group with double-dose buddleja officinalis extract(D) and therapy group with genistein(E).The dry eye model was established with ORX on Group B,C,D,E.Group C,D,E were gastrically perfused with single-dose or double-does buddleja officinalis extract or genistein for 30 days.All rabbits were detected with SIT.TGF-β1,IL-1β,TNF-αwere detected on immunohistochemistry.Results:The SIT value of group C,D,E is significantly higher than that of group B(P〈0.01).The expression of IL-1β,TNF-αin acinar cell and glandular tube cell of group C,D,E were significantly higher than that of group B(P〈0.01) and the expression of TGF-β1 in acinar cell and glandular tube cell of group C,D,E were significantly higher than that of group B too(P〈0.01).Any result of group C,D was positive than that of group E(P〈0.05).Conclusion:The main components of extract of buddleja officinalis is the flavonoids.The flavonoids can displays androgen-like activity.So it can adjust endogenous gonadal hormone level.As a result,it can adjust lacrimal gland partial inflammation.Extract of buddleja officinalis may be an effective therapeutics to this kind of dry eye because of its characteristic.
