摘 要:目的:分析中药复方"松友饮"及组方药材提取物指纹图谱用于质量控制。方法:采用高效液相色谱法(high-performance liquid chromatography,HPLC)研究中药指纹图谱。包括低压四元泵,自动进样器和二极管阵列检测器(DAD),Agilent Zorbax RX-C8色谱柱(4.6 mm×250 mm,5μm),柱温25℃,流动相为水-乙腈,进行梯度洗脱(流速,0.5 mL/min;洗脱时间,60 min;检测波长,280nm)。结果:成功构建"松友饮"及组方提取物高效液相指纹图谱,色谱保留时间稳定,各主要色谱峰偏移小于0.1 min。通过比对发现"松友饮"HPLC色谱峰来自组方药材成分。重复HPLC色谱相似度(90%,精密度、稳定性、重复性实验中各共有峰相对保留时间和相对峰面积RSD(3%。结论:HPLC方法可靠、可行,指纹图谱重复性良好,为"松友饮"质控提供参考。Objective: To analyze the fingerprints of Chinese herbal extracts of compound "Songyou Yin" and its components for quality control.Methods: High performance liquid chromatography(HPLC) technique was applied to study fingerprints of the herbs.The mobile phase consisted of acetonitrile and water was used as gradient elution for HPLC separation,which was performed on Agilent Zorbax RX C8(4.6mm×250mm,5μm) chromatographic column,including low pressure quaternionic pump,autosampler,and detected at 280 nm with a DAD detector.The flow rate was 0.5 mL/min,proceeding time was 60 min,and the column temperature was set at 25℃.Results: The fingerprints,with relative stable retention time,were successfully established for extracts of "Songyou Yin" and its components by HPLC technique.The differences in principal fingerprint peaks were less than 0.1 min.The fingerprint peaks of "Songyou Yin" derived from those of components(Astragalus membranaceus Bge.,Salvia miltiorrhiza Bge.,Lycium barbarum L.and Crataegus pinnatiWda Bge.) showed a greater than 90% similarity in repeated experiments.The RSD of the relative retention time as well as that of the relative peak area of the shared peak was lower than 3% in the precision,stability and reproducibility tests.Conclusion: The fingerprints established by HPLC method were reliable,feasible,and reproducible for the quality control and assessment of Chinese herbal compound "Songyou Yin".
