摘 要:目的:探讨护津方对细菌脂多糖(Lipopolysaccharide,LPS)诱导的急性肺损伤大鼠支气管肺泡灌洗液(Bronchoalveolar lavage fluid,BALF)中弹性蛋白酶(Neutrophil elastase,NE)及肺组织肿瘤坏死因子-αmRNA(Tumor necrosis factor-α mRNA,TNF-α mRNA)表达的影响。方法:40只雄性Wistar大鼠随机分为5组:正常对照组、LPS模型组、地塞米松组、护津方组,每组再分为4h和8h两个亚组。分别用地塞米松和护津方在LPS诱导ALI模型前预处理大鼠。分别采用酶联免疫吸附法和实时荧光定量PCR检测NE及TNF-α mRNA的表达。结果:两个时相点中,与模型组相比,护津方组BALF中NE含量均显著降低(P<0.01)。与模型组相比,8h组中,护津方组TNF-α mRNA表达均显著降低(P<0.01)。病理学观察,模型组大鼠肺组织出现大片出血及坏死,护津方组大鼠肺组织病理损伤程度较模型组减轻。结论:护津方能减轻内毒素致急性肺损伤大鼠肺组织损伤,对内毒素致急性肺损伤大鼠有保护作用,其机理可能与其能降低内毒素致急性肺损伤BALF中NE及肺组织TNF-α mRNA的表达有关。Objective:To investigate the effects of Hujin Decoction on the changes of expression of Neutrophil elastase(NE) in Bronchoalveolar lavage fluid(BALF) and Tumor necrosis factor-α mRNA(TNF-α mRNA) in the rats caused by LPS.Methods:Forty male Wistar rats were randomly divided into 4 groups: normal control group,LPS model group,dexamethasone group and Hujin Decoction group.Each group had two subgroups,4h and 8h after LPS was injected.All rats,except normal control group,were administrated with LPS by intravenous injection to induce acute lung injury.The rats in dexamethasone group and Hujin Decoction group were treated by dexamethasone(oral),Hujin Decoction(oral) respectively before LPS-induced ALI.The animals were killed after injection of LPS for 4 and 8 hours.Then the expression of NE and TNF-αRNA were measured by ELISA and real-time fluorescent PCR while the histopathology of the lung injury was observed by light microscope.Results:In both 4h and 8h groups,compared with model group,the expression of NE in BALF in Hujin Decoction group was obviously decreased(P〈0.01).Compared with model group,the expression of TNF-α mRNA in 8h groups was obviously decreased in Hujin Decoction group(P〈0.01).Pathological observation shows that pulmonary hemorrhage and necrosis were largely seen in model group and a better condition in Hujin Decoction group.Conclusion: Hujin Decoction may abate the histopathology of the lung injury and they had protective effects on endotoxin-induced acute lung injury in rats,and the drugs might take effect by means of inhibiting the expression of NE in BALF and TNF-αRNA in lung tissues.
