摘  要:目的:观察葛根素对新发现的小分子活性肽Apelin在大鼠心肌缺血/再灌注损伤中表达的影响。方法:将SD大鼠60只,随机分成4组:缺血/再灌注(IR)组即缺血20min后再灌注180min;葛根素+缺血/再灌注(IP)组;假手术(SH)组和正常对照(NC)组。体表心电图标Ⅱ导联全程记录,进行心律失常得分评估;用放免法测定各组大鼠血浆中Apelin-36蛋白含量变化;RT-PCR方法观察心肌Apelin mRNA表达。结果:(1)葛根素IP组心律失常得分仅为IR组的69.9%(P<0.01)。(2)血浆Apelin-36浓度:SH组、IR组、IP组分别是NC组93.4%(P>0.05)、51.5%(P<0.01)和73.5%(P<0.01)。IP组比IR组含量高,两者有统计学差异(P<0.01)。(3)心肌Apelin mRNA的表达:IR组明显降低,仅为NC组的40.2%(P<0.01);IP组为NC组的68.1%(P<0.01);IP组比IR组高(P<0.05)。结论:药物葛根素处理后能使大鼠血浆Apelin-36含量及心肌中ApelinmRNA表达升高。提示葛根素能影响小分子活性肽Apelin在大鼠心肌缺血/再灌注损伤过程中的表达。Objective:To observe the effects of Puerarin on expression of Apelin at the myocardial ischemia reperfusion injury rats.Methods:Sixty adult healthy male Sprague-Dawley(SD) rats were randomly divided into ischemia reperfusion injury(IR) group,puerarin preconditioning(IP)group,normal control(NC) group,and left coronary artery not was tighted in(SH) group.A standard limb lead ⅡECG was continuously monitored throughout the experiment to record all kinds of arrhythmias to evaluate the score of arrhythmias.The plasma Apelin-36 were detected by radiommunoassay.The expression of Apelin mRNA was investigated in left ventricle myocardium by RT-PCR.Results:The scores of arrhythm,IP were 69.9% of IR group(P〈0.01).(2)The plasma Apelin-36 in SH group was 93.4% of NC group(P〈0.05);IR group was 51.5% of NC(P〈0.01).IP group was 73.5% of NC group(P〈0.01);The plasma Apelin-36 in IP group was higher than in IR group(P〈0.01).(3)Compared with NC group,the expression of Apelin mRNA in left ventricle myocardium vitally reduced in IR group,and was only 40.2% of NC group(P〈0.01);The level of expression in IP group was 68.1% of NC group(P〈0.01);The expression of Apelin mRNA in IP group increased compared with IR group(P〈0.05).Conclusion:The plasma Apelin-36 concentration and the expression of Apelin mRNA in myocardium are significantly higher at Puerarin preconditioning group in pathogenesis of the myocardial ischemia reperfusion injury.These changes indicate that Puerarin have the effects on expression of Apelin at the myocardial ischemia reperfusion injury rats.
