管藻目绿藻叶绿素蛋白复合物PAGE分离方法的优化

摘  要:以高等植物菠菜为对照,对管藻目绿藻刺松藻和假根羽藻叶绿素蛋白复合物的PAGE分离方法进行了优化.选择SDS,DOC,DMG和TritonX-1004种去污剂在不同的浓度、比例及增溶时间条件下,对色素蛋白复合物的分离效果进行了比较,并对电泳分离条件做了部分改进,提高了分辨率.采用改进后的方法从刺松藻和假根羽藻中各分离到11种色素蛋白复合物,除文献中报道的复合物外,还分离到两种新的捕光复合物Fca1和Fca2.该方法为进一步研究管藻目绿藻色素蛋白复合物的特性提供了条件.The conditions of detergent resolve and PAGE separation in the isolation of chlorophyll protein complexes of siphonous green algae are optimized and compared with those of spinach. The separation effects of chlorophyll protein complexes resolved by different kinds of detergents which are SDS, DOC, DMG, Triton X 100, at different concentration, weight ratio and reaction time are compared. And the separation conditions are partially improved. By the new method with higher resolution, 11 kinds of pigment protein complexes are resolved from thylakoiks of C. fragile and B. Corticulans, respectively. Two of them named Fca 1 and Fca 2 are light harvesting chlorophyll protein copmlexes which have never been reported before. Here we offer a good method for further study of chlorophyll protein complexes of siphonous green algae.
