摘  要:利用全基因组基因芯片技术对0.2Gyγ射线照后2、12、24小时点的人淋巴细胞和未照射的人淋巴细胞基因差异表达谱进行了研究和分析。结果表明,照后2小时的差异表达基因有44条,其中明显上调的差异表达基因有38条,明显下调的差异表达基因6条;照后12小时的差异表达基因有247条,其中明显上调的差异表达基因有151条,明显下调的差异表达基因96条;照后24小时的差异表达基因有704条差异基因表达谱,其中明显上调的差异表达基因有392条,明显下调的差异表达基因312条;共同差异表达基因只有6条。主要涉及到以下几类型基因:干扰素调节有关基因;生长抑制、DNA损伤有关基因;与P53调控有关的基因;与肿瘤坏死因子有关的基因;与细胞周期调控及凋亡有关的基因等等。RT-PCR结果表明,MERTK及TAIAP12个差异表达基因,其相对定量结果与基因芯片检验结果相一致。The study analyzed the differential transcriptional profile of normal human lymphocyte and human lymphocyte irradiated with 0.2 Gy γ ray by whole genome chip after 2,12,24 h point irradiated.The results showed that there were 44 differentially expressed genes after 2 hour irradiated,of which 38 genes were up-regulated and 6 genes down-regulated.The results showed that there were 247 differentially expressed genes after 12 hour irradiated,of which 151 genes were up-regulated and 96 genes down-regulated.While the results showed that there were 704 differentially expressed genes after 24 hour irradiated,of which 392 genes were up-regulated and 312 genes down-regulated.These genes are associated with interferon-regulated genes,GADD45,P53-regulated genes,tumor necrosis factor etc.RT-PCR results indicated that the relative quantitative result of MERTK and TAIAP1 are consistent with gene chip data.
