摘  要:目的对pERK在幼鼠睡眠剥夺所致的学习记忆障碍中的作用进行初步探讨,以明确睡眠剥夺对儿童学习记忆的影响及其信号机制。方法 1月龄幼鼠分为正常对照组、大平台对照组、睡眠剥夺组,使用睡眠剥夺箱建立睡眠剥夺模型,使用Morris水迷宫训练检测幼鼠学习与记忆能力,使用免疫组织化学染色方法检测水迷宫测试后幼鼠海马CA1、CA2区pERK1/2表达水平。结果睡眠剥夺组幼鼠CA1、CA2脑区pERK1/2表达则明显少于两对照组(P值均小于0.01),且着色浅淡。训练第1天,各组平均潜伏期无明显差异,但从第2天开始,睡眠剥夺组幼鼠的平台潜伏期较其它两个对照组出现延长(P<0.05),而且在最后1天进行的空间探索试验中,睡眠剥夺组幼鼠穿环数与正常及大平台对照组相比均明显减少(P<0.01)。结论 幼鼠海马容易受到睡眠剥夺的影响,从而导致空间记忆的破坏和海马ERK磷酸化的减弱。同时也说明海马ERK激活的下降参与了睡眠剥夺导致的空间记忆的破坏。Objective To investigate the role of pERK on learning and memory impairment induced by sleep deprivation(SD)in young rats,and to clarify the effect of sleep deprivation on children's learning and memory and its signaling mechanism.Methods 1 month-aged rats were divided into normal control group(NC),big penal group(BPC)and sleep deprivation group(SD).Sleep deprivation box were used to make SD model.Morris water maze were used for the testing of rats' learning and memory ability,by using immunohistochemistry method to detect the expression of pERK1/2 in CA1,CA2 area of hippocampus.Results pERK1/2 expression in CA1,CA2 area of SD group was significantly less and weaker than the other two control groups(P〈0.01).At the 1st day of Morris water maze training,every group had no significant difference in average panel latency(APL)(P〉0.05).But from the 2nd day,SD group had longer APL than the other two groups(P〈0.05).And in the Spatial probe test at the last day,SD group had less times of passing through the platform(P〈0.01).Conclusions The hippocampus of young rats can be easily affected by SD.SD can result to the destruction of space memory and the reduced expression of pERK.
