摘 要:目的:探讨过度训练及补充谷氨酰胺对大鼠海马和额叶皮质中TNF-α及其mRNA和NF-κB水平的影响。方法:60只SD大鼠随机分为3组,安静组(C,n=20)、过度训练组(E,n=20)和谷氨酰胺补充组(G,n=20),E组和G组进行递增负荷跑台训练11周,最后一次训练结束后36 h断头取海马和额叶皮质。应用ELISA方法检测TNF-α的蛋白含量,Real ti me-PCR方法检测TNF-α的mRNA转录,Western blot检测NF-κB的蛋白表达。结果:1)与C组比较,E组海马和额叶皮质中TNF-α蛋白含量显著升高(分别为P<0.01和P<0.05);与E组比较,G组海马中TNF-α蛋白含量显著降低(P<0.01),额叶皮质中未见显著性变化(P>0.05);2)与C组比较,E组海马和额叶皮质中TNF-αmRNA的表达水平显著升高(均为P<0.05);与E组比较,G组海马中TNF-αmRNA的表达水平显著下降(P<0.05),额叶皮质中未见显著性变化(P>0.05);3)与C组比较,E组海马和额叶皮质中NF-κB的表达水平显著升高(P<0.05);与E组比较,G组海马和额叶皮质中NF-κB的表达水平均未见显著性变化(均为P>0.05)。结论:1)过度训练可以明显影响大鼠海马和额叶皮质中TNF-α及其mRNA和NF-κB的表达水平;2)谷氨酰胺补充可以在一定程度上逆转大鼠过度训练时海马中TNF-α及其mRNA的表达水平,但对NF-κB的表达不会产生影响。Objective: To investigate the effects of overtraining and pre-glutamine supplementation on TNF-α, its mRNA and NF-κB in hippocampus and frontal cortex of rat brain. Methods : 60 male sprague-dawley rats were randomly divided into three groups : blank control group (C, n = 20 ), overtraining group ( E, n = 20 ) and glutamine supplementation group ( G, n = 20). E and G group were training in standard treadmill with an increasing load for 11 weeks. 36h after the last treadmill training, all rats were sacrificed by decapitation. ELISA, real time- PCR and western blot were used to detect the expression of TNF-α, its mRNA and NF-κB respectively. Results: 1. Compared with C group, the level of TNF-a was increased both in hippocampus and frontal cortex in E group. The level of TNF-a was decreased in hippocampus in G group compared with V. group. 2. The expression of TNF-α mRNA were up-regulated in hippocampus and frontal cortex in E group compared with C group, and only down-regulated in hippocampus in G group compared with E group. 3. Compared with C group, the expression of NF-κB was increased in hippocampus and frontal cortex in E group and compared with E group, the expression of NF-κB were not changed both in hippocampus and frontal cortex in G group. Conclusion.. 1. Overtraining could impact the expression of TNF-a, its mRNA and NF- κB in hippocampus and frontal cortex of rat brain. 2. Pre-glutamine supplementation could reverse the expression of TNF-α and its mRNA and in hippocampus of overtraining rat brain partly, hut could not affect the expression of NF-κB.
