摘 要:目的:探讨Sarcopenia关联的线粒体介导的细胞凋亡信号通路中各凋亡基因的增龄变化情况及爬梯运动对其的影响。方法:以快速老化(Senescence-accelerated mice prone/8,SAMP8)小鼠为衰老动物模型,将其分为青年安静组(YC组)、老年安静组(OC组)和老年爬梯运动组(OR组),采用实时荧光定量PCR的方法,分别对各组腓肠肌Caspase依赖性凋亡通路中相关基因(Cyt C、apaf-1、Caspase-9和Caspase-3)和Caspase非依赖性凋亡通路中相关基因(PARP、AIF和Endo G)进行检测。结果:OC组腓肠肌Sarcopenia Index(SI)值显著低于YC组;随增龄OC组腓肠肌中apaf-1、Caspase-3和PARP的表达均显著上调,而AIF却显著下调;爬梯运动能够下调OR组腓肠肌中Cyt C和Caspase-3的表达,而AIF和Endo G却未见显著性变化。结论:老年SAMP8小鼠腓肠肌可作为Sarcopenia研究模型;Sarcopenia关联的线粒体介导的细胞凋亡信号通路中Caspase依赖与Caspase非依赖两条凋亡途径发挥的作用存在一定差异,衰老骨骼肌质量衰减与否以及衰减程度最终有赖于两条途径之间的对抗;8周爬梯运动能够一定程度上弱化老年小鼠腓肠肌中的凋亡信号,避免其更大范围地进入凋亡程序而加剧Sarcopenia,对"脆弱"的骨骼肌起到一定的保护作用。Objective : To discuss the change of apoptotic genes in Sarcopenia related mitoehondria mediated apoptotic signaling pathway, and the effects of ladder climbing exercise on it. Methods:SAMP8 mice are selected as the aging model animal, with the grouping of young control group (YC),old control group (OC) and old resistant exercise group (OR). The method of real-time PCR is used to detect the changes of apoptotic genes, such as Cyt C, apaf-1, Caspase- 9, Caspase-3, PARP, AIF and Endo G in each group. Results: (1) Value of SI in gastroenemius of OC group is lower than that of YC group significantly; (2) Expression of apaf-1, Caspase-3 and PARP increase significantly with aging, on the contrary the expression of AIF decreases significantly; (3) Ladder climbing exercise down-regulates the expression of Cyt C and Caspase-3, but has no effect on AIF and Endo G. Conclusion: (1) The gastrocnernius of senile SAMP8 mice can be used as a model for the research of Sarcopenia; (2) Sarcopenia related mitochondrial mediated Caspase dependent and independent apoptotic signaling pathways show different directions during apoptotic selection, therefore, the final result, Sarcopenia or not, depends on the antagonism between the two pathways; (3) Eight weeks ladder climbing can attenuate the inclination of the apoptosis in the frail senile mice to avoid further advancing for apoptosis and Sarcopenia finally.
