摘  要:目的:研究玉竹多糖(PoPs)预处理对实验性糖尿病大鼠胰岛β细胞损伤的影响。方法:给PoPs灌胃预处理正常大鼠,28d后按180mg·kg-1一次性腹腔注射四氧嘧啶诱导实验性糖尿病,检测造模后大鼠的FBG、INS、MDA、SOD、GSH-Px、CAT的水平,取胰腺组织做HE染色观察胰岛损伤情况。结果:四氧嘧啶诱导的实验性糖尿病大鼠胰岛细胞存在明显的氧化应激损伤。PoPs预处理呈计量依赖性地降低糖尿病大鼠FBG,使胰腺组织MDA含量减少,SOD、GSH-Px、CAT活力增强,HE染色显示胰岛损伤明显减轻。结论:玉竹多糖具有降血糖、抗氧化应激作用,可以明显减轻四氧嘧啶对糖尿病大鼠胰岛β细胞的损伤。OBJECTIVE To evaluate the possible protective effects and mechanism of pretreatment with Polygonatum odoraturn polysaceharides (PoPs) against β-cell damage in experimental diabetes in rats. METHODS Pretreating with PoPs by oral administration in normal rats, 28 day later, alloxan was injected intraperitoneally at a single dose of 180 mg· kg^-1 for induction of diabetes. The level of body weight,FBG, INS in serum and MDA, SOD, GSH-Px and CAT contents in pancreatic homogenates were assayed by spectrophotometry. Pancreas samples were stained with HE and examined under a light microscope. RESULTS Alloxan successfully induced experimental diabetes in rats, pancreatic island significantly damaged by oxidative stress. Pretreatment with PoPs decreased fasting blood glucose levels and blocked the increase of MDA production and increased SOD.GSH-Px.CAT activity in pancreatic homogenates as a dose dependent manner compared to the diabetic control(P〈0.05). Histopathological examination showed that the size and cell number of islets were reduced in diabetic control group and PoPs restored the damage of pancreas tissues in rats with diabetes mellitus. CONCLUSION Pretreatment with PoPs has protective effect on pancreatic β-cell damage associated with hypoglycemic effect and reduction of oxidative stress in alloxan-induced diabetes in rats.
