摘 要:为了培育生长速度快的牙鲆养殖品种,以缩短育种周期,提高牙鲆养殖产品的质量和经济效益,利用微卫星标记技术对牙鲆亲本遗传距离与后代生长速度间的相关性进行了分析研究。选用21个多态性微卫星分子标记,以8个日本牙鲆(JS)个体和24个抗病牙鲆(RS)个体为亲本产生的28个杂交组合为材料,分析了该32个牙鲆亲本的遗传多样性,同时对牙鲆亲本间遗传距离与其后代生长速度的相关性进行了研究。结果表明:(1)21个微卫星标记共扩增出123个等位基因,32个牙鲆亲本中的等位基因数(A),有效等位基因数(Ae),观测杂合度(Ho),期望杂合度(He),多态信息含量(PIC)分别在2~15,1.3423~7.8168,0.2333~1.000,0.2593~0.8859,0.2225~0.8613之间变动,相应的平均值分别为5.86,3.7951,0.6931,0.6784,0.6237。(2)进行亲本间遗传距离(GD)与其后代生长速度(AGRw)的相关性分析发现,亲本间最小遗传距离为0.2578,最大遗传距离为0.9773,不同遗传距离范围内,亲本间遗传距离与后代生长速度之间呈现不同的相关性:在0.2578~0.5958范围内,亲本间遗传距离与后代生长速度呈显著的正相关(P<0.05);在0.6099~0.6604范围内,亲本间遗传距离与后代生长速度之间没有表现出显著的相关性;在0.6640~0.9773范围内,亲本间遗传距离与后代生长速度呈显著的负相关(P<0.05)。To cultivate the aquaculture species of flounder with rapid growing rate,microsatellite marker technology was used to analyze the correlation between parental genetic distance and the growth traits of their offspring.It is of great significance for shortening the breeding cycle,and improving the quality and economic benefit of artificial farmed flounder.Eight Japanese flounders (JS) and 24 resistant flounders (RS) were used to make 28 crosses.The genetic diversity of these 32 parents was analyzed using 21 polymorphic microsatellite markers.Simultaneously,the correlation between parent SSR genetic distance and the growth rate of their offspring was assayed.The results indicated that:(1) the number of total alleles of these 21 microsatellite loci was 123,the allele number (A),effective allele number (A e),observed heterozygosity (H o),expectedheterozygosity (H e) and polymorphism information content (PIC),ranged from 2-15,1.3423~7.8168,0.2333-1.000,0.2593-0.8859,0.2225-0.8613,respectively.The average A,A e,H o,H e,PIC was 5.86,3.7951,0.6931,0.6784,0.6237,respectively.(2) The minimum genetic distance was 0.2578,while the maximum was 0.9773.The correlation between AGRw of offspring and parental GD existed considerable difference in different range of genetic distance.As the GD for 0.2578-0.5958,the correlation between AGRw of offspring and parental GD appeared to be positively significant at P 0.05 ;As the GD for 0.6099~0.6604,there was no significant correlation ;Furthermore,as the GD for 0.6640-0.9773,the correlation between AGRw of offspring and parental GD appeared to be negatively significant at P 0.05.
