摘  要:目的研究白细胞介素-8(IL-8)对血管内皮细胞通透性的影响。方法采用Transwell弥散模型,以标志物漏出法检测IL-8不同浓度、不同作用时间对EA.hy926细胞单层通透性的影响;结晶紫染色法检测细胞形态学改变;透射电镜进行细胞连接形态学观察。结果IL-8可明显增加血管内皮细胞单层的通透性(P〈0.05),呈一定的剂量和时间依赖关系;荧光倒置相差显微镜观察,随着IL-8浓度和作用时间的增加,细胞明显回缩,细胞间隙明显增加;透射电镜实验表明,随着IL-8浓度和作用时间的增加。细胞连接逐渐打开、破坏。结论IL-8以时间和剂量依赖方式引起血管内皮细胞通透性升高、细胞及细胞连接形态学的改变。Objective To explore the influence of IL-8 on the permeability of vascular endothelial cells. Methods Vascular endothelial cells were cultured on the inserts of transwell filters. At different times after the addition of IL-8 of different concentration to the upper wells, the alternation of cell permeability was measured. Crystal violet staining was used to observe the morphological change of vascular endothelial cells. Transmission electron microscope was used to observe the morphology of the cell-cell junctions. Results The results demonstrated that IL-8 induced increase in the permeability of vascular endothelial cells in a dose and time dependent manner. Fluorescence inverted microscopic results showed that vascular endothelial cells recovered and the intercellular space increased with the increase of the dose of IL-8 or culture time. TEM observation indicated that the cell-cell junctions were opened and destroyed gradually with the increase of dose or culture time of IL-8. Conclusion IL-8 induces the increase of the permeability of vascular endothelial cells, alters the morphology of vascular endothelial cells, and destroys the cell-cell junctions in a dose and time dependent manner.
