摘  要:目的观察在大鼠诱发性肝癌中有无血管生成拟态(VM)现象,并研究其与肝癌分化程度、侵袭转移的相关性。方法建立二乙基亚硝胺(DEN)和N-亚硝基吗啉(NMOR)联合诱导的具有转移潜能的♂SD大鼠肝癌模型,应用HE染色、免疫组化、透射电子显微镜等手段描述VM现象。结果成功构建了具有肝内转移播散的♂SD大鼠诱发性肝癌动物模型,在肝癌模型标本中观察到VM现象。分化级别低的肝癌组织比分化级别高的肝癌组织中VM现象多见,两组阳性率分别为61.3%、8.3%,差异有统计学意义(χ2=9.752,P<0.05);在有肝内播散转移的肝癌组织标本中VM现象较无转移的肝癌组织标本多见,VM阳性率分别为60.7%、2.0%,差异有统计学意义(χ2=6.508,P<0.05)。结论在DEN联合NMOR诱导的♂SD大鼠肝癌模型中存在VM现象,该现象与诱发性肝癌分化程度、肝内侵袭转移相关。Objective To observe the relationships between the formation of vasulogenic mimicry(VM)and differentiation grade,invasion and metastasis,and study a rat model of chemically induced hepatocellular carcinoma(HCC).Methods Set up the male SD rat model with metastatic potential of diethylnitrosamine(DEN)and N-nitrosomorpholine (NMOR) induced HCC.Used methods included hematoxylin-eosin staining,immunohistochemistry,transmission electron microscope to detect the formation of vasculogenic mimicry.Results This study successfully constructed the male SD rat model with intrahepatic metastasis of DEN and NMOR induced HCC.We successfully detected the phenomena of vasculagenic mimicry in HCC.The phenomena of VM were related to the HCC differentiation grade,lower grade HCC had more VM(VM positive rate 61.3% vs 8.3%,χ^2=9.752,P〈0.05).The phenomena of VM were related to the HCC invasion and intrahepatic metastasis,HCC with more invasion and metastasis had more VM (VM positive rate 60.7% vs 2.0%,χ^2=6.508,P〈0.05).Conclusion The phenomena of VM are related to the HCC differentiation grade,invasion and intrahepatic metastasis,in the male SD rat model,DEN and NMOR induced.
