摘 要:目的:探讨双氢青蒿素对早期放射性肺损伤大鼠肺组织及血清肿瘤坏死因子α(TNF-α)表达的动态影响。方法:采用γ射线单次全肺照射15Gy,建立SD大鼠放射性肺损伤模型。108只同体质量雄性SD大鼠,随机分为1)正常对照组:灌服等体积溶媒;2)单纯照射组:灌服等体积溶媒+全肺单次照射15Gy;3)照射用药组:灌服双氢青蒿素+全肺单次照射15Gy。双氢青蒿素和溶媒皆于照前3d开始灌服持续至照射后15d。ELSIA法及免疫组化方法检测照射后60d内血清及肺组织TNF-α表达情况。结果:单纯照射组肺组织炎症渗出及纤维化征象严重。照射15d,单纯照射组、照射用药组肺组织及血清TNF-α表达依次递增,单纯照射组肺组织TNF-α表达最强,照射用药组介于正常对照组和单纯照射组之间(P<0.05),照射后30d,两组差异无统计学意义(P>0.05)。血清TNF-α两组表现为从照射后3d开始增加,到15d达到高峰,30d与正常对照组无差别,照射用药组较单纯照射组明显减少(P<0.05)。结论:双氢青蒿素具有降低早期放射性肺损伤大鼠血清、肺组织TNF-α表达的作用。OBJECTIVE:To investigate the ability of dihydroartemisinin to affect TNF-α releasing in the animal model,so as to find an effective method in reducing the lung toxicity after thoracic irradiation.METHODS:The chest of 108 SD rats were exposed to either sham irradiation or single fraction irradiation of 15 Gy after having been randomized into 3 groups:NT group received drenching same volume solventia;XRT group received whole lung 15 Gy irradiation and drenched the same volume solventia without dihydroartemisinin;DA/XRT group received both drenching dihydroartemisinin and whole lung 15 Gy irradiation.Drenching once a day began from 3 days before radiation to 15 days after radiation.Elisa and SP method were used to check the expression of TNF-α in blood serum.RESULTS:The inflammation and fibrosis of the XRT group was the most severe,after radiation fifteen days after radiation,the NT group exhibited low level of TNF-α protein expression.There was a significantly elevated level of TNF-α positive inflammatory cells in the XRT group.The gray scale of the positive cells in the DA/XRT group was between the NT and XRT group with a significant difference(P〈0.05).Thirty days after radiation,the expression of TNF-α in rat lung was not different(P〉0.05)between the DA/XRT group and XRT group.The expression change of TNF-α in blood serum was the same as in lung tissue in the 3rd and 15th day after radiation.In the 30th day,there was difference between DA/XRT and XRT groups(P〈0.05).CONCLUSION:Dihydroartemisinin can restrain the expression of TNF-α in blood serum and lung tissue,which is possible the mechanism.
