摘 要:目的探讨SH2-B在结肠癌癌变过程中的演变规律,分析SH2-B表达与结肠癌临床分期的关系,分析SH2-B表达与结肠癌转移的关系。方法免疫组织化学方法检测69例结肠癌组织、27例结肠腺瘤组织、29例结肠息肉组织中SH2-B蛋白的表达。69例结肠癌患者Ⅰ期11例、Ⅱ期22例、Ⅲ期19例、Ⅳ期17例,其中无转移34例,淋巴节转移35例。结果免疫组织化学检测结果显示,SH2-B蛋白在结肠息肉组织、结肠腺瘤组织、结肠癌组织中的表达呈浆-核型,以胞浆为主。结肠息肉患者SH2-B阳性率为31%(9/29),结肠腺瘤患者SH2-B阳性率为52%(14/27),结肠癌患者SH2-B阳性率为93%(27/29)。结肠癌组织中SH2-B的阳性细胞率(0.64±0.26)明显高于结肠腺瘤组织(0.26±0.27)和结肠息肉组织(0.12±0.21)(P<0.001),结肠腺瘤组织中阳性细胞表达率明显高于结肠息肉组织(P<0.05)。结肠癌Ⅰ、Ⅱ、Ⅲ和Ⅳ期强阳性率分别为18.2%(2/11)、45.4%(10/22)、57.9%(11/19)和82.3%(14/17)。转移组和非转移组强阳性率为45.7%(16/35)和23.5%(8/34),转移组SH2-B强阳性率显著高于无转移组(P<0.05)。结论 SH2-B过表达可能参与了结肠癌癌变过程,可能是结肠癌变的早期事件。SH2-B表达与结肠临床分期有关。SH2-B可能参与了结肠癌转移,SH2-B高表达与结肠癌转移呈正相关。[ Objective] To investigate the expression profile of SH2-B in colon carcinogenesis and analyze the relationship of SH2-B expression and color cancer chnical stages or metastasis. [ Methods ] Using immunohistoehemistry assay, SH2-B expression was detected in 69 eases of colon cancer, including 11 eases of stage Ⅰ, 22 of stage 11, 19 of stage Ⅲ and 17 of stage Ⅳ, 27 of colonic adenomas, 29 of colonic polyps and 38 of nasopharyngeal carcinoma. The relationship of SH2-B expression and colon cancer chnie was analyzed. [Results] SH2-B expressed in cytoplast and cytoplasm of colonic polyps, colonic adenoma and colon cancer tissues,mainly located in cytoplasm. The positive rate was 31% in colon polyps (9/29), 52% in colonic adenoma (14/27), and 93% in colon cancer tissue (27/29). Positive cell rate in colon cancer tissues (0.64±0.26) was significantly higher than that in colon polyps (0.12±0.21) and colon adenoma tissues (0.26±0.27) (P 〈0.001),colonic adenomas was significantly higher than colon polyps (P 〈0.05). The positive rates of stage Ⅰ, Ⅱ, Ⅲ, Ⅳ were 18.2 % (2/11), 45.4 % (10/22), 57.9% (11/19) and 82.3% (14/17), respectively. SH2-B gradually increased along with stage Ⅰ ,Ⅱ, Ⅲ, Ⅳ. SH2-B expression was associated with clinic stages. The positive rates of metastasis and no metastasis were respectively 23.5% (8/34) and 45.7% (16/35), the positive rate was higher in metastasis than no metastasis (P 〈0.05). [Conclusion] SH2-B ex- pression gradually increased in colon carcinogenesis. This implies that SH2-B over-expression is an early event. It is also associated with colon cancer clinical stages, and might involve in colon cancer metastasis.
