padi4-104单核苷酸多态性与河北省汉族人群类风湿关节炎的相关性研究

摘　　要：目的探讨中国河北汉族人群肽酰基精氨酸脱亚胺酶4-104(padi4-104)单核苷酸多态性(SNP)与RA易感性的关系。方法用PCR-DNA测序法对无血缘关系的115名RA患者和106名健康体检者的PADI4基因padi4-104位点的SNP进行检测分析。结果 padi4-104的基因型分布符合Hardy-Weinberg平衡(P>0.05),AA、AG、GG基因型和A、G等位基因分布在RA病例组与对照组间差异均无统计学意义(P值分别为0.368、0.245、0.252),比值比(OR)分别为0.709(95%可信区间:0.336～1.498)、0.636(95%可信区间:0.296～1.364)、0.800(95%可信区间:0.546～1.172)。结论河北汉族人群PADI4基因的padi4-104位点具有多态性,但此单核苷酸多态性与RA的易感性无相关性。Objective To study the association of peptidylrginine deiminase 4-104 （padi4-104） genetic single nucleotide polymorphism（SNP） and rheumatoid arthritis susceptibility in Han population in Hebei province. Methods The genotypes and allele frequencies of PADI4 gene polymorphisms （padi4-104） were analyzed by PCR-DNA sequencing method, Results The distribution of padi4-104 genotypes between the two groups was not significantly deviate from that expected by Hardy-Weinberg equilibrium （P〉0.05）, the alleles distribution of the padi4-104 between the two groups was not significantly different, and OR was 0.709（95% CI： 0.336-1.498）, 0.636 （95%CI： 0.296-1.364）, 0.800 （95%CI： 0.546~1.172）, respectively. Genotypes distribution and alleles frequencies of padi4-104 didn＇t show notable difference between Han population and Japanese, Spain（P〉0.05）. But the alleles frequencies of padi4-104 was significantly different between Hebei province eproviand UK（P=0.039）. Conclusion In Han population of Hebei, the SNP of padi4-104 is not associated with the incidence of RA.
