黄连、吴茱萸及其药对对DMH诱导的大鼠结肠肠腺增殖与凋亡的影响

摘　　要：目的:研究黄连、吴茱萸及其药对大鼠结肠肠腺细胞增殖与凋亡的影响。方法:皮下注射二甲肼(1,2-dimethyl-hydrazine、DMH)诱发大鼠结肠ACF(异常腺窝病灶,Aberrant Crypt Foci,ACF)形成,观察Fulgen染色法后结肠肠腺细胞增殖和凋亡的变化。结果:黄连具有抑制细胞增殖作用,吴茱萸及黄连吴茱萸药对具有抑制细胞增殖、促进凋亡作用,且药对效果优于单味药。结论:黄连、吴茱萸及其药对具有潜在的抑制结直肠腺癌的作用可能与其促进肠腺细胞凋亡和抑制增殖相关;黄连吴茱萸配伍药对优于单味药。Objective：To study Influence of huanglian,wuzhuyu and huanglian-wuzhuyu pair herb on colon cell proliferation and apoptosis of rat induced by DMH（1,2-dimethylhydrazine）.Methods：DMH （1,2-dimethylhydrazine） was injected subcutaneously to induce Aberrant Crypt Foci （ACF） in rat colon.The frequencies of mitotic and apoptotic cells within intact microdissected crypts was examined after stained with Fulgen reagent.Results：Cell proliferation was inhibited by huanglian;cell proliferation was inhibited and apoptosis was promoted by wuzhuyu and huanglian-wuzhuyu pair herb,and herb pair effect was better than single herb.Conclusion：Huanglian,wuzhuyu and huanglian-wuzhuyu pair herb may be potential chemoprevention agents against colorectal cancer and this effect may be associated,at least in part,with inhibition of cell mitosis and promotion of cell apoptosis;pair herbs composed by huanglian-wuzhuyu may have better effect than single herb.
