摘  要:为了研究翼手目序列中马铁菊头蝠DRB基因第2外显子的遗传多态现象,试验检测了从一个种群得到的17个蝙蝠个体。结果从17个个体中克隆获得42条不同序列,长度为263 bp(仅4例为260 bp)。经序列分析发现,部分个体存有多种序列、不同个体间存有相同序列,提示马铁菊头蝠MHCⅡ-DRB基因第2外显子可能存在着重复座位和基因共享。氨基酸的替换趋于集中在假定的抗原结合位点附近。利用MEGA软件构建的NJ树表明,马铁菊头蝠聚为一支,而与其他动物分离。结果表明:马铁菊头蝠MHCⅡ-DRB基因第2外显子有较高的多态性,这与其他哺乳动物很相似。During the process of order Chiroptera study, genetic diversity of exon 2 in the Rhinolophus ferrumequinum was discovered. 17 bats in the same population was detected in this assay, and 42 different sequences whose length is all 263 bp (only with 4 samples are 260 bp) were obtained by sequencing. Sequence analysis showed that more than two sequences were found in the same individual and vice versa, suggesting that there would be locus duplication and gene sharing in the class II major histocompatibility complex DRB gene of Rhinolophus ferrumequinum. The substitutions of amino acid tended to gather around the putative antigen-binding sites. The neighbor-joining(NJ)method constructed by MEGA soft-ware indicated that Rhinolophus ferrumequinum were the same species, different from other species. This study showed that the high level of highpolymorphism existed in the exon 2 of the MHC class Ⅱ gene DRB of Rhinolophus ferrumequinum, which was similar to other mammals.
