摘 要:目的探讨雷公藤单体T4对胰岛β细胞的免疫保护作用,为胰岛β细胞的免疫保护治疗提供实验依据。方法培养胰岛β细胞株NIT细胞及正常对照外周血单个核细胞,分别与以下6组作用:①IL-1β+IFN-γ组;②IL-1β+IFN-γ+DXM组;③IL-1β+IFN-γ+T4组;④T4组;⑤DXM组;⑥对照组(DMEM)。ELISA法检测培养上清IL-4、IFN-γ水平,硝酸还原酶法检测上清一氧化氮水平,化学发光法检测上清胰岛素水平。结果①IL-1β+IFN-γ刺激的NIT-1细胞分泌IL-4水平较其他各组减少(P<0.01),IFN-γ分泌比其他各组增多(P<0.01),IL-4/IFN-γ比其他各组降低(P<0.01)。②T4+IL-1β+IFN-γ刺激的NIT-1细胞与IL-1β+IFN-γ刺激组相比,分泌IL-4水平增高(P<0.01),IFN-γ减少(P<0.01),IL-4/IFN-γ增高(P<0.01)。③IL-1β+IFN-γ刺激的NIT-1细胞分泌NO较其他各组增高(P<0.01),胰岛素分泌减少(vsIL-1β+IFN-γ+T4组、IL-1β+IFN-γ+DXM组,P<0.05;vsT4组、DXM组、对照组,P<0.01)。④T4+IL-1β+IFN-γ刺激的NIT-1细胞与IL-1β+IFN-γ组相比,分泌NO水平减少(P<0.01),Ins增加(P<0.05);与IL-1β+IFN-γ+DXM组相比,NO分泌减少(P<0.05)。结论 T4可减轻IL-1β+IFN-γ对NIT细胞的损伤作用,保护NIT细胞的胰岛素分泌功能。Objective To investigate the protective mechanism of tripchlorolide on islet β cells and to provide experiment evidence of using tripchlorolide for immunoprotection of islet β cells.Methods NIT Islet β cell line and peripheral blood mononuclear cells from the control subjects were treated with (1) IL-1β+IFN-γ,(2) IL-1β+IFNγ+DXM,(3) IL-1β+IFN-γ+T4,(4) T4,(5) DXM or (6) DMEM.The levels of IL-4 and IFN-γ in the culture supernatants were measured with enzyme-linked immunosorbent assay (ELISA).Nitric oxide was measured with nitrate reductase assay and the level of insulin was tested with chemiluminescence assay.Results ①After treatment with IL1β+IFN-γ,the concentration of IL-4 in the culture supernatants was lower than the other groups (P〈0.01),while the level of IFN-γ was higher(P〈0.01) than the other groups.The IL-4/IFN-γ was also lower(P〈0.01) than other goups.②After treatment with T4+IL-1β+IFN-γ,the concentration of IL-4 in the culture supernatants was higher than IL-1β+IFN-γ group (P〈0.01) while the level of IFN-γ was lower (P〈0.01) and IL-4/IFN-γ was higher (P〈0.01) than IL-1β+IFN-γ goup.③In addition to the cytokines,the concentration of NO in supernatants of IL-1β+IFN-γ stimulation group was higher than the other groups (P〈0.01) while insulin level was lower than the other groups(vs IL-1β+IFN-γ+T4 group,IL-1β+IFN-γ+DXM group,P〈0.05;vs T4 group,DXM group and DMEM control,P 〈0.01).④The concentration of NO in supernatants of T4+IL-1β+IFN-γ stimulation group was lower than IL-1β+IFN-γ group (P〈0.01) and IL-1β+IFN-γ+DXM group (P〈0.05) while insulin level was higher than IL-1β+IFN-γ group(P〈0.05).Conclusion T4 could alleviate the damage by the combined action of IL-1β +IFN-γ on NIT cells and maintain the insulin secretion function of NIT cells.
