摘 要:目的为了探讨bcl 2基因在系统性红斑狼疮(SLE)发病机制中的作用及临床意义。方法应用逆转录 -聚合酶链反应(RT PCR)检测31例SLE患者外周血单一核细胞(PBMC)bcl 2mRNA表达水平和流式细胞仪双标记法分析其T、B细胞bcl 2蛋白表达。结果活动期SLE患者PBMCbcl 2mRNA表达水平明显升高 ,占55 6 % ,且活动期SLE患者CD3+ 、CD4+ 和CD8+ T细胞亚群bcl 2蛋白表达明显高于非活动期SLE患者、正常人对照组和其他疾病组 ,而CD19+ B细胞bcl 2蛋白表达在各组之间并无统计学差异。结论T细胞bcl 2异常表达在SLE疾病活动的过程中起着重要作用 ,检测SLE患者外周血T淋巴细胞bclObjective To investigate the role of bcl 2 expression in the pathogenesis of systemic lupus erythematosus (SLE) and its clinical significance Methods The bcl 2 mRNA levels in peripheral blood mononuclear cells (PBMC) from 31 SLE patients were tested by reverse transcription polymerase chain reaction (RT PCR), and the bcl 2 protein levels in T and B cells were also tested by two colour cytofluorography Results bcl 2 mRNA levels in PBMC increased significantly in 10 cases of active SLE, accounting for 55 6% of active SLE patients Compared with those in inactive SLE patients, normal controls and other non SLE patients, the bcl 2 protein expression in T cell subsets (including CD3+, CD4+ and CD8+ ) increased significantly in active SLE patients However, there was no statistical difference in bcl 2 protein levels in CD19+ B cells among all groups Conclusion The abnormal expression of apoptosis related bcl 2 in T cells might play an important role in disease activity of SLE , and the determination of bcl 2 expression levels of peripheral T lymphocytes might be used as an index for disease activity of SLE
