金叶败毒制剂对人巨细胞病毒感染宿主细胞周期的调节作用

摘　　要：目的:探讨中药金叶败毒制剂(简称中药)对人巨细胞病毒(HCMV)感染宿主细胞周期的影响。方法:用HCMV AD169感染人胚肺成纤维细胞(HEL),用金叶败毒制剂干预后,采用免疫组织化学法检测HCMV感染及中药干预后宿主细胞P16蛋白的表达,并采用流式细胞技术检测HCMV感染及药物干预后宿主细胞周期。结果:在HCMV感染48h后,金叶败毒制剂能明显抑制宿主细胞P16蛋白的表达。经中药干预后,在病毒感染24h后,宿主细胞处于S期细胞明显减少;病毒感染48h后,宿主细胞处于G2/M期的细胞明显增多;在HCMV感染72h后,宿主细胞均表现为S期细胞明显减少,G2/M期细胞明显增多,而G0/G1期细胞亦明显增多。结论:金叶败毒制剂可能通过抑制HCMV感染细胞P16蛋白的表达,促进感染细胞从S期进展到G2/M期,促进细胞周期进程而发挥抗病毒作用。Objective：To explore the effect of Jinye Baidu preparation （JBP） on cell cycle of human cytomegalovirus （HCMV） infected host cells.Methods：Human embryonic fibroblasts were infected by HCMV AD169,then intervened by Jinye Baidu preparation,the expression of P16 protein in host cells after HCMV infection and chinese herbs intervention was detected by immunohistochemistry; the cell cycle of host cells after HCMV infection and chinese herbs intervention was detected by flow cytometry.Results：48 hours after HCMV infection,Jinye Baidu preparation inhibited the expression of P16 protein in host cells significantly,after chinese herbs intervention,the amount of host cells at S phase decreased significantly after virus infection for 24 hours,while the amount of host cells at G2/M phase increased significantly after virus infection for 48 hours; the amount of host cells at S phase decreased significantly and the amounts of host cells at G2/M phase and G0/G1 phase increased significantly after virus infection for 72 hours.Conclusion：Jinye Baidu preparation can inhibit the expression of P16 protein in host cells after HCMV infection,advance the infected cells at S phase to G2/M phase,promote the process of cell cycle so as to achieve the antiviral effect.
