摘  要:目的观察姜黄素对亚硒酸钠诱导的体外培养兔晶状体混浊的影响,探讨姜黄素对抗晶状体氧化损伤的效果。方法将体外培养的40只正常家兔的晶状体分为5组:空白对照组、亚硒酸钠组(30μmol/L)、姜黄素A组(30mg/L)、姜黄素B组(50mg/L)及卡林-U组(质量分数1%)。分别于培养后24、48、72、96h观察各组晶状体的透明度,同等条件下用数码相机拍照记录各组晶状体的形态,用分光光度计测定晶状体组织中的谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、超氧化物歧化酶(SOD)的变化。结果在48h时,不同质量分数姜黄素组与空白对照组比较,不同分级的晶状体混浊的眼数差异无统计学意义(H=1.464,P>0.05),姜黄素组与卡林-U组及亚硒酸钠组比较,不同分级的晶状体混浊眼数的差异有统计学意义(H=10.607,P<0.01);72h时,不同质量分数姜黄素组与空白对照组比较,不同分级的晶状体混浊的眼数差异无统计学意义(H=3.288,P>0.05);姜黄素组与卡林-U组及亚硒酸钠组比较,不同分级的晶状体混浊眼数的差异有统计学意义(H=8.258,P<0.05)。晶状体培养后96h,不同质量分数姜黄素组与空白对照组比较,不同分级的晶状体混浊眼数的差异有统计学意义(U=7.102,P<0.05);不同质量分数姜黄素组与卡林-U组及亚硒酸钠组比较,不同分级的晶状体混浊眼数的差异有统计学意义(H=9.699,P<0.05)。与空白对照组比较,亚硒酸钠组GSH-Px、SOD活性明显降低,MDA浓度明显升高。不同质量分数姜黄素组和卡林-U组的GSH-Px、SOD活性高于亚硒酸钠组,低于空白对照组,MDA浓度介于空白对照组与亚硒酸钠组之间,卡林-U组的GSH-Px、SOD活性低于姜黄素组,MDA浓度高于姜黄素组。结论姜黄素及卡林-U均具有抗氧化损伤作用,均可推迟亚硒酸钠诱导的体外培养兔晶状体混浊的发生时间,姜黄素的作用强于卡林-U,但姜黄素与卡林-U最终均不能抑制白内障的发生。Background Experimental research demonstrated that oxidative damage was associated with age-related cataract closely.Curcumin has the effect of anti-inflammatory,anti oxidation,anti tumor and the role of immune suppression.Objective The purpose of this research was to assess the effect of curcumin on sodium selenite-induced cataract.Methods The lenses were obtained from 40 rabbits and cultured in DMEM containing 10% fetal bovine serum and randomly divided into 5 groups:control group,sodium selenite-treated group (30 μmol/L ),30 mg/L curcumin-treated group,50 mg/L curcumin-treated group and catalin-treated group (1%).The transparence of lenses cultured in vitro were observed and graded in the time points of 24,48,72 and 96 hours.At the end of the incubation period,the contents of glutathion peroxidase(GSH-Px),malondialdehyde (MDA) and superoxide dismutase (SOD) in lenses were examined for biochemical parameters by spectrophotometry.Results No obvious difference was found in the numbers of opacity lens between curcumin-treated lenses and control group (P〉0.05),but significant difference was seen between the curcumin-treated group and sodium selenite-treated group or catalin-treated group in 48 hours and 72 hours after culture(H=10.607,P〈0.01;H=8.258,P〈0.05).In the time point of 96 hours,the numbers of opacity lens were considerably decreased in curcumin-treated group compared with control group (u=7.102,P〈0.05),catalin-treated group and sodium selenite-treated group (H=9.699,P〈0.05).The content of MDA was evidently increased,and those of GSH-Px and SOD were significantly decreased in sodium selenite-treated group compared with control group (P〈0.01).The contents of GSH-Px and SOD in lens in curcumin-treated group and catalin group were higher than that of the sodium selenite-treated group and were lower than that of the control group(P〈0.01).The contents of MDA in lens in different curcumin-treated group and catalin-treated group were lower than that of t
