摘　　要：目的寻找自然流产模型小鼠(CBA/J×DBA/2)子宫蜕膜组织与正常妊娠小鼠(CBA/J×BALB/c)子宫蜕膜组织之间差异表达的miRNAs。方法建立自然流产小鼠模型和正常妊娠小鼠模型,利用miRCURY LNATMmicroRNAArrays 10.0检测两组孕小鼠子宫蜕膜组织miRNAs的表达,筛选两组小鼠子宫蜕膜组织差异表达的miRNAs,并对其进行功能检索和靶基因预测。结果 CBA/J×DBA/2小鼠胚胎吸收率为30.9%,CBA/J×BALB/c小鼠胚胎吸收率为7.0%,两者差异有统计学意义(P<0.01)。miRNAs芯片数据表明流产蜕膜组织和正常蜕膜组织之间有13个miRNAs存在明显表达差异(ratios≥2.0或≤0.5),其中表达上调8个,表达下调5个。预测软件显示miR-21和miR-26a的靶基因分别是和妊娠相关的RECK和LIF两个因子。结论小鼠子宫蜕膜组织miR-21和miR-26a的差异性表达可能在小鼠胚胎自然流产过程中扮演重要角色。Objective To observe the differential expression of microRNAs in decidua of spontaneous abortion mice （CBA/J ×DBA/2） and normal pregnant mice （CBA/J ×BALB/e）. Methods A spontaneous abortion mouse model and a normal pregnant mouse model were established. Expression of miRNAs in decidua of the two models was detected by miRCURY LNATM nricroRNA array 10. 0. Differential expression of microRNAs was detected by function analysis and prediction of target genes. Results The embryo resorption rate was significantly higher in the CBA/J x DBA/2 group than in the CBA/J × BALB/c group （30.9% vs 7.0% ,P 〈 0.01 ）. The data on microRNA chip showed that the expression of 13 miRNAs, including up-regulated and down-regulated expression of 8 and 5 miRNAs （ ratios ≥ 2.0 or ≤ 0.5 ） was significantly different in decidual tissue of spontaneous abortion and normal pregnant mice. The prediction software showed that the target genes of miR-26a and miR-21 were LIF and RECK. Conclusion Differential expression of miR-26a and miR-21 in decidua may play an important role in spontaneous abortion of mice.
