补肺益肾方对幼龄哮喘豚鼠肺内嗜酸粒细胞凋亡和IL-5受体表达的影响

摘　　要：目的观察中药补肺益肾方对幼龄哮喘豚鼠支气管肺泡灌洗液(bronchoalveo larlavage fluid,BALF)中嗜酸粒细胞(eosinophils,EOS)白介素-5受体α(interleukin-5Rα,IL-5Rα)mRNA表达的影响,探讨中药补肺益肾方促进哮喘EOS凋亡的机制。方法 24只健康幼龄豚鼠随机分为正常组、模型组、中药组。以卵蛋白致敏激发制备豚鼠哮喘模型,分离豚鼠BALF中的EOS,TdT介导的dUTP缺口末端标记法(TdT-mediated dUTP nick end labeling ,TUNEL)检测细胞凋亡,原位杂交检测IL-5RαmRNA表达。结果模型组豚鼠EOS凋亡明显减少,中药组EOS凋亡明显高于模型组(P<0.01)。模型组豚鼠EOS表达IL-5RαmRNA明显低于正常组(P<0.01),中药组EOSIL-5RαmRNA表达明显增加(P<0.05)。结论中药补肺益肾方可通过促进IL-5RαmRNA表达,降低IL-5生物学功能,促进EOS凋亡,减轻气道炎症,从而达到防治哮喘的目的 。Objective The purpose of this study was to investigate the effect of the Chinese medicinal recipe Bufei Yishen （invigorating the lung and nourishing the kidney） on the expression of IL-5 receptor α mRNA in the bronchoalveolar lavage fluid （BALF） eosinophils of young guinea pigs with asthma,and to explore the mechanism of the recipe promoting eosinophil apoptosis.Methods Twenty-four young guinea pigs were randomly divided into three groups,normal control asthma model,and Bufei Yishen.The asthma models were established by ovalbumin sensitization,and eosinophils were purified from BALF.The apoptosis of the eosinophils was detected by TdT-mediated dUTP nick end labeling （TUNEL）,and the expression of IL-5 IL-5Rα mRNA measured by in situ hybridization.Results The apoptosis of eosinophils was significantly lower in the models than in the Bufei Yishen group （P〈0.01）.The expression of IL-5Rα mRNA in eosinophils was markedly increased in the Bufei Yishen group （P〈0.05）,but it was significantly lower in the models than in the normal controls （P〈0.01）.Conclusion The Chinese medicinal recipe Bufei Yishen promotes the apoptosis of eosinophils by increasing the expression of IL-5 R α mRNA and decreasing the effect of IL-5,and thus contributes to the alleviation of airway inflammation and prevention of asthma.
