摘 要:采用免疫组织化学方法研究了Caspase-3和Bcl-2蛋白在7日龄、14日龄、28日龄和56日龄小鼠睾丸组织的表达,旨在观察小鼠出生后睾丸发育和精子发生过程中细胞凋亡相关蛋白Caspase-3和Bcl-2的定位。结果发现Caspase-3在7日龄、14日龄小鼠睾丸组织表达于曲精小管中央和管周生精细胞,在28日龄和56日龄表达于管周精原细胞与支持细胞,初级精母细胞及以后各级生精细胞无Caspase-3表达;Bcl-2在7日龄、14日龄睾丸组织表达于曲精小管中央和管周生精细胞,28日龄和56日龄广泛表达于初级精母细胞以后各级生精细胞,精原细胞选择性表达Bcl-2。Caspase-3和Bcl-2在小鼠睾丸组织发育和精子发生过程中表达。在小鼠出生后睾丸发育的早期,Caspase-3和Bcl-2表达于曲精小管中央,参与曲精小管管腔形成的调节;在性成熟后的睾丸组织中Caspase-3表达于精原细胞,诱导其凋亡;Bcl-2于初级精母细胞以后各级生精细胞高表达,抑制精母以后生殖细胞的凋亡。The present study investigated the expression of Caspase-3 and Bcl-2 in mouse testis during the process of postnatal development and sperm occurrence of the pastnatal mouse. The expression of Caspase- 3 and Bel-2 in the testis of 7day old,14 day old,28 day old and 56 day old mouse were observed by the immunohistochemical method. The results showed that Caspase-3 and Bcl-2 expressed in the testis of all stages were observed. Caspase-3 positive cells were observed in the central part of seminiferous and the spermatogonia of 7-day-old and 14-day-old mouse's testis,in the spermatogonia and Sertoli cells of 28-day-old and 56-day-old mouse's testis. No Caspase-3 positive was observed in the spermatocytes and spermatozoa. In the testis of 7-day-old and 14-day-old mouse,Bcl-2 localized in central part and spermatogonia. In the testis of 28-day-old and 56-day-old mouse, Bcl-2 localized in the seminiferous compartment,with a preferential adluminal compartment staining in the seminiferous tubules. Caspase-3 and Bcl-2 expressed in the testis of mouse during the postnatal development and spermoccurrence. During the early stage of postnatal development of mouse testis,Caspase-3 and Bel-2 localized in the central part of seminiferous, which indicated that Caspase-3 and Bcl-2 involved in the process of lumen's formation of seminiferous. In the process of spermoccurrence,Caspase-3 mainly occurred in spermatogonia, which indicated that Caspase-3 involved in the regulation of apoptotic process of spermatogonia Bcl-2 might inhibit the germ cell death of postspermatocyte.
