摘 要:目的:检测变应性鼻炎患者IL-9的表达水平,探讨IL-9在变应性鼻炎发生发展中的意义。方法:收集49例变应性鼻炎患者的外周血标本,其中未治疗组30例,临床缓解组19例,另取30例健康志愿者的外周血标本作为正常对照组。采用实时荧光定量PCR(QRT-PCR)检测IL-9基因在外周血中的表达水平,酶联免疫吸附试验(ELISA)检测血浆IL-9含量。结果:变应性鼻炎未治疗组外周血单个核细胞中IL-9 mRNA表达水平及血浆IL-9含量明显高于变应性鼻炎临床缓解组和正常对照组(P<0.05),而变应性鼻炎临床缓解组与正常对照组比较差异无统计学意义。变应性鼻炎未治疗组IL-9 mRNA表达水平与临床评分呈正相关。结论:变应性鼻炎患者外周血单个核细胞中IL-9呈高表达。IL-9可能参与了变应性鼻炎的病理过程,具体机制有待进一步探讨。Objective: To establish a method of real-time fluorescence quantitative PCR for detecting IL-9 mRNA in human peripheral blood mononuclear cell (PBMC) , and investigate the relationship between the expression of IL-9 and allergic rhinitis (AR). Methods: The mRNA levels of IL-9 in PBMC were determined by quantitative real-time PCR (QRT-PCR) , and detected the concentrations of IL-9 in plasma by enzyme linked immunosorbent assay (ELISA) in 49 patients, including 30 untreated AR patients and 19 patients in recovering phase, and 30 healthy control volunteers. Results: IL-9 mRNA expression and IL-9 concentrations in the group of 30 AR patients without treatment were markedly higher than that in the groups of 19 AR patients in clinical remission and 30 healthy controls ( P 〈 0.05 ). but there were no significant differences between the groups of AR in clinical remission and normal control (P 〉 0.05 ). Furthermore, the result of this study shows a positively correlated between IL-9 mRNA and clinical count in untreated AR patients. Conclusion: These results indicated that IL-9 was higher in AR patients without treatment, and suggesting that IL-9 might be involved in the pathological process of AR. The role of IL-9 in process of AR remains further investigation.
