摘  要:目的:探讨腺病毒载体介导的绿色荧光蛋白基因转染大鼠骨髓间质干细胞(mesenchymal stem cells,MSCs)的生物学特性。方法:骨髓贴壁法分离培养大鼠骨髓MSCs并采用流式细胞术进行鉴定。利用脂质体法通过腺病毒转染获得EGFP修饰的大鼠骨髓MSCs。结果:成功分离得到大鼠MSCs,腺病毒转染后大鼠骨髓MSCs过表达绿色荧光蛋白(EGFP)报告基因,且转染后MSCs具有成骨分化潜能。结论:通过腺病毒转染能获得EGFP标记的大鼠MSCs,为MSCs组织工程和基因治疗研究提供了依据。Objective: To investigate the biological characteristics of rat bone marrow mesenchymal stem cells (MSCs) transfected green fluorescent protein(GFP) gene by adenovirus vector. Methods: Rat bone marrow-derived MSCs were isolated by adherent culture method. Flow cytometric analyses were used to identify cells which were transfected by recombinant adenovirus-EGFP using lipofectamine2000. Results: Rat bone marrow-derived MSCs were successfully isolated and efficiently express EGFP report gene by adenovirus vectors transfection. Conclusion: Rat bone marrow-derived MSCs can be transfected by recombinant adenovirus-EGFP, which may potentially be used as a target for tissue engineering and gene therapy.
