异丙嗪对下丘脑细胞内游离钙浓度的影响

摘　　要：目的：观察异丙嗪（ｐｒｏｍｅｔｈａｚｉｎｅ，ＰＭＺ）对下丘脑细胞内游离钙浓度（［Ｃａ２＋］ｉ）的影响。方法：以酶法制备家兔下丘脑细胞悬液，运用钙指示剂Ｆｕｒａ－２／ＡＭ作为细胞内游离钙的荧光探针测定下丘脑细胞［Ｃａ２＋］ｉ。结果：１）ＰＭＺ（０４６ｍｍｏｌ／Ｌ）使下丘脑［Ｃａ２＋］ｉ显著升高，且在一定的剂量范围内呈量效关系。２）事先向细胞悬液中加入钙通道阻滞剂维拉帕米可明显抑制ＰＭＺ诱导的［Ｃａ２＋］ｉ升高，但不能完全阻断ＰＭＺ的这种作用。结论：上述结果提示ＰＭＺ可引起下丘脑［Ｃａ２＋］ｉ升高，钙通道开放导致细胞外钙内流是ＰＭＺ引起下丘脑［Ｃａ２＋］ｉ升高的机制之一。The metabolism of active sludge carbohydrate is inve The Effect of Promethazine on Free Intracellular Calcium Concentration in Dissociated Hypothalamus Cells$$$$ Yan Yuxia 1, Wang Huadong 2, Wang Yanping 2, Chen Xiaolin 2 1Dept. of Biochemistry; 2 Dept.of Pathophysiology, JNUMC, Guangzhou 510632 China Abstract: Aim: The purpose of this study was to examine the effect of promethazine(PMZ) on free intracellular Ca 2+ concentrations([Ca 2+ ] i) in freshly dissociated hypothalamus cells isolated from New Zealand rabbits.Methods:[Ca 2+ ] i was measured by Fura-2 fluorometry.Results:PMZ (0.46 mmol/L) induced a significant increase of [Ca 2+ ] i,pretreatment with Ca 2+ channel blocker verapamil(0.28 mmol/L) significantly inhibited the elevation of [Ca 2+ ] i induced by PMZ,but verapamil couldn't completely blocked the PMZ-induced increase of [Ca 2+ ] i.Conclusion:These results indicate that a increase of [Ca 2+ ] i induced by PMZ is partly dependent on influx of Ca 2+ from the extracellular space through Ca 2+ channel.
