摘 要:水稻基因RID1突变后表现为"不开花"的表型,且RID1特异在幼叶中表达。本研究利用启动子RPP16,构建表达载体RPP16::RID1,转化rid1突变体,得到了19株转基因阳性植株,其中有9株表现为恢复抽穗,表明在维管组织中特异表达RID1可以恢复rid1突变体表型。RT-PCR检测结果显示,转基因单株中抽穗期基因Ehd1、Hd3a和RFT1的表达量与RID1表达量呈正相关,且RID1表达量高的转基因单株相应抽穗也比较早。The RID1 gene expressed in immature leaf of rice especially.When the RID1 gene is knocked out,the rice never headed and thus referred to as a never-flowering phenotype.We constucted a expressed vector named RPP16::RID1 by employing the promoter RPP16 in this study.And the vector RPP16::RID1 was transformed into rid1 mutant background by Agrobacterium-mediated transformation,9 out of 19 positive transgenic plants are flowering normally,which indicated the RID1 gene expressed specially in rice phloem tissues can complement the phenotype of rid1 mutant.The results of RT-PCR demonstrated RID1 gene regulated the expression of Ehd1,Hd3a,RFT1 positively,and the higher RID1 expression,the earlier the corresponding plant flowered.
