摘  要:本研究以南紫薇、福建紫薇以及37个栽培品种为材料,利用AFLP分子标记分析其亲缘关系。PCR实验结果表明,清晰计数的条带共有270条,多态性条带占了220条,百分比最高为80.98%;单对引物组合扩增出的可计数条带最高81条,最少32条,五对引物组合扩增出的多态性条带平均54条,多态性百分比最高为84.5%,最低为74.47%;种质资源相似系数为0.5407~0.9151,平均相似系数为0.6846。UPGMA聚类结果显示39个材料可划分成三大部分,其中南紫薇及南紫薇与紫薇产生的杂交品种被划分为第Ⅰ组,福建紫薇及福建紫薇与紫薇产生的杂交品种划分为第Ⅱ组,所有紫薇种间的杂交品种划分到第Ⅲ组。本实验表明AFLP标记技术能从分子水平上较为准确地体现紫薇种质资源的亲缘关系。The genetic relationship of Lagerstroemia subcostata,Lagerstroemia limii and 37 cultivated varieties was analyzed by AFLP markers.Result of PCR experiment showed that among the 270 polymorphic bands obtained from 5 selective primer pairs,220 was polymorphic,accounting for 80.98%.The average number of DNA bands per primer pair was 54 ranging from 32 to 81,with a maximum polymorphic rate of 84.5% and a minimum rate of 74.47%.The genetic similarity among all 39 accessions evaluated ranged from 0.540 7 to 0.915 1,with an average of 0.684 6.Based on the UPGMA dendrogram,all the materials could be divided into three types.The first category included Lagerstroemia subcostata together with the hybrids of Lagerstroemia subcostata and Lagerstroemia;The second category included Lagerstroemia limii as well as the hybrids of Lagerstroemia limi and Lagerstroemia;The third category included all the hybrids between Lagerstroemia indica cultivars.This study revealed that AFLP approach was efficient in generating accurate information on the genetic diversity and relationship of Lagerstroemia germplasm resources.
