摘 要:目的:研究转录调节因子RUNX1在银屑病患者骨髓CD34+细胞中的表达,以揭示银屑病患者造血干细胞的活性。方法:采用免疫磁珠法分离CD34^+细胞,流式细胞术鉴定细胞纯度,反转录PCR和蛋白免疫印迹法分别检测RUNX1的mRNA和蛋白表达。结果:银屑病患者骨髓CD34^+细胞RUNX1 mRNA及蛋白表达水平均高于正常对照组(P<0.05)。结论:决定CD34^+细胞发育分化的关键转录因子RUNX1在银屑病患者骨髓CD34^+细胞中表达异常,RUNX1表达水平的增高可能引起骨髓造血干细胞和造血微环境的异常,并可能参与银屑病的发病。Objective:To reveal the activity of hematopoietic cells and examine the expression of RUNX1 in bone marrow CD34^+ cells from patients with psoriasis. Methods: Bone marrow CD34^+ cells were isolated from psoriatic patients and normal persons by immunomagnetic cell selection, purity of cells was evaluated by flow cytometry. The expression of mRNA and protein for RUNX1 was analyzed using reverse transcriptase-polymerase chain reaction (RT-PCR) and Western-blot. Results: The expression of RUNX1 in bone marrow CD34^+ cells from patients with psoriasis was significantly higher than that from normal controls (P〈0.05). Conclusions: The genes were abnormal, which influences the differentiation of bone marrow CD34^+ cells from patients with psoriasis. The abnormality of RUNX1 in bone marrow of psoriasis perhaps plays a role in the pathogenesis of psoriasis.
