CD40-CD40L在寻常性银屑病患者外周血的表达及意义

摘　　要：目的:探讨单核细胞上CD40和其配体CD40L及血清中游离型配体sCD40L在寻常性银屑病患者外周血中的水平及其临床意义。方法:采用流式细胞术检测30例寻常性银屑病患者外周血单核细胞CD40、CD40L表达水平,并采用双抗体夹心酶联免疫吸附法(ELISA)检测血清sCD40L的浓度。另取20例正常人外周血作为对照组。结果:寻常性银屑病患者外周血单核细胞上CD40的表达水平及血清中sCD40L浓度明显高于对照组(P<0.01),外周血单核细胞CD40与CD40L表达水平呈正相关(r=0.412,P<0.05),外周血单核细胞CD40表达水平与血清中sCD40L浓度呈正相关(r=0.487,P<0.05)。结论:CD40-CD40L通路在银屑病患者免疫功能紊乱中起了重要作用。Objective： To investigate the level of the expression of CD40-CD40L in human peripheral blood mononuclear cell and serum soluble CD40L（sCD40L） in patients with psoriasis vulgaris and its significance. Methods： The positivity rates of CD40 and CD40L in human leucocytes were measured by flow eytometry in 30 patients with psoriasis vulgaris. Levels of sCD40L in sera were measured by double antibody sandwich enzyme-linked immunosorbent assay （ELISA）. Results： The positivity rates of CD40 in human leucocytes and the serum levels of sCD40L were significantly increased in patients with psoriasis vulgaris as compared with normal controls （P〈 0.01 ）, and the expression of CD40 was correlated positively with CD40L（r=0.412,P〈0,05） and the concentration of sCD40L in sera（r= 0.487 ,P〈0.05）. Conclusions： CD40-CD40L pathway may play important roles in the pathogenesis of psoriasis vulgaris, is possible to he a good therapeutic target for the treatment of psoriasis.
