摘 要:抗菌肽是一类具有抗菌活性的多肽类物质。利用农杆菌介导法将中国对虾抗菌肽基因np3和np5分别导入粳稻品种爱知旭中,PCR检测显示,T0代转np3基因和np5基因水稻植株的阳性率分别为36%和45%。RT-PCR检测T1代转基因水稻,结果表明抗菌肽基因在RNA水平上得到表达,而抗菌肽的表达对农艺性状未产生显著影响。用白叶枯病菌小种CR4分别对4个T1代转np3基因和np5基因株系进行接种,所有转基因株系均表现出比对照更高水平的抗性。为了验证抗菌肽基因的广谱抗性,进一步检测了np5转基因株系对3个菌株JS97-2、ZHE173和OS-225的抗性,发现中国对虾抗菌肽基因np5对白叶枯病菌可能具有较广谱的抗性。Antimicrobial peptides (AMP) is a polypeptide with antimicrobial activity.The antimicrobial peptide genes np3 and np5 from Chinese shrimp were integrated into Oryza sativa L.subsp.japonica.cv.Aichi ashahi by Agrobacterium mediated transformation system.PCR analysis showed that the positive ratios of np3 and np5 were 36% and 45% in T0 generation,respectively.RT-PCR analysis showed that the antimicrobial peptide genes were expressed in T1 generation and there was no obvious difference in agronomic traits between transgenic plants and non-transgenic plants.Four np3 and np5 transgenic lines in T1 generation were inoculated with the strain CR4 of Xanthomonas oryzae pv.oryzae(Xoo),and the results indicated that all the transgenic lines had a significantly enhanced resistance to bacterial blight caused by the Xoo strain CR4.The np5 transgenic lines also conferred higher resistance to bacterial blight caused by the Xoo strains JS97-2,ZHE173 and OS-225.It is suggested that transgenic lines with np5 gene might possess a broad spectrum resistance to rice bacterial blight.
