摘 要:目的:动态观察大鼠糖尿病肾病发生发展过程中转化生长因子(TGF)-β1的下游正负反馈调节因子Smad7、Psmad2在肾脏的表达变化及对细胞外基质主要成分CollagenⅣ表达的影响。方法:40只SD大鼠随机分成4组:CON、DM4W、DM8W、DM16W组。STZ诱导糖尿病肾病模型,Masson染色动态观察各组大鼠胶原合成变化。Westernblot、免疫组化方法动态观察Smad7、Psmad2、CollagenⅣ表达变化。结果:Masson染色结果示,随糖尿病肾病进展,肾内胶原生成逐渐增多,Smad7从4周开始即比正常组表达减少,且随时间进展逐渐减少(P<0.05)。Psmad2、CollagenⅣ从4周开始表达增多,随时间的延长逐渐增多(P<0.05)。到16周Smad7/Psmad2明显失衡。结论:TGF-β/Smad信号通路的正负反馈调节蛋白失衡导致细胞外基质大量沉积及胶原生成可能是糖尿病肾病肾脏纤维化进展重要信号转导途径之一。Objective To study the dynamic changes of expression of Psmad2 and Smad7 in TGF-β/Smad pathway and their effects on the expression of CollagenIV, a main component of extracellular matrix in the development of renal fibrosis in diabetic nephropathy. Methods Thirty Sprague-Dawley rats injected with STZ were randomly divided into 4W,8W and 16W groups, and another ten normal rats were used as negative control group. The dynamic change of collagen was assessed by Masson staining. And the expression of Smad7 and CollagenⅣ were analyzed by Western blotting and immunohistochemistry respectively. Results Masson staining reveals that collagen production increased with the progression of diabetic nephropathy. While the expression of Smad7 reduced with the progression of nephropathy, the expression of Psamd2 and Collageniv increased. Compared to the control group, the expression of those three genes showed statistically significant differences in group DM4W, DM8W and DM16W respectively. And the imbalance between the expression of Smad7 and Psmad2 is obvious after 16 weeks. Conclusion The deposition of extracellular matrix and collagen caused by the imbalanced expression of the positive and negative feedback regulators in TGF-β/Smads pathway may be one of the most important signal transduction pathways in the development of diabetic nephropathy.
