摘 要:采用农杆菌介导的遗传转化法对玉米强致病性弯孢菌CX-3进行转化,得到具潮霉素B抗性且能稳定遗传的转化子30个。随机选取11个突变子进行PCR鉴定,结果表明外源T-DNA片段已经整合到CX-3的基因组中。通过形态学观察发现6个转化子在颜色、生长速度及产孢量方面有明显变化。Agrobacterium tumefaciens-mediated transformation (ATMT) was a highly efficient transformation to filamentous fungi,it has been extensively applied to many fungi, such as Fusarium, Trichoderma,etc.Curvularia lunata CX-3, which was strong pathogenicity to maize, andit was successfully transformed by ATMT, 30 genetically stable transformants was obtained, and all of them had resistance to hygromycin B.11 of them were randomly selected to conduct PCR, and the result showed that the T-DNA had been integrated into genome of CX-3.Compared to wild strain CX-3,6 transformants were obviously different in colour, growth rate and spore production.
