碘缺乏和甲状腺功能减退对大鼠仔鼠内嗅皮质CaMKⅡ表达的影响

摘  要:目的观察碘缺乏和甲状腺功能减退损伤内嗅皮质对钙调蛋白依赖性蛋白激酶(calcium/calmodulin pro-tein kinase II,CaMKⅡ)表达的影响。方法健康2月龄雌性Wistar大鼠,交配妊娠后,取孕鼠28只,按体重随机分成对照组、甲状腺功能减退组和碘缺乏组,甲状腺功能减退组根据饮水中含丙基硫尿嘧啶(propylthiouracil,PTU)剂量分为5ppm组和15 ppm组,每组7只孕鼠。分别于PN7、PN14、PN21、PN28和PN42时,每组随机取5只仔鼠,灌流固定大脑,进行组织病理切片和免疫组化染色,观察分析内嗅皮质CaMKⅡ表达。结果PN7,PN14和PN21时,15 ppm组和碘缺乏组仔鼠内嗅皮质CaMKⅡ的表达显著低于对照组(P<0.05),PN28和PN42时,5 ppm组、15 ppm组和碘缺乏组仔鼠内嗅皮质CaMKⅡ的表达亦显著低于对照组(P<0.05)。结论碘缺乏和甲状腺功能减退可降低内嗅皮质CaMKⅡ表达。Objective To observe the effect of iodine deficiency and hypothyroidism on the protein expressions of CaMKⅡ in the entorhinal cortex of pups.Methods Female Wistar rats(n=28) after pregenancy were randomly divided into control group,hypothyroid group and iodine deficient group.According to the dose of PTU in fed water,hypothyroid group was divided into 5ppm group and 15ppm group(7 in each).5 pups from each group were sacrificed and perfused intracardially in PN7,PN14,PN21,PN28 and PN42.Brains were removed,fixed and sectioned coronally.All sections were observed and analysed the protein exression of CaMKⅡ by immunohistochemistry in the entorhinal cortex.Results In PN7,PN14 and PN21,protein levels of CaMKⅡ in the entorhinal cortex in iodine-deficient and 15ppm treatment groups was significantly lower than those of controls(P〈0.05).In PN28 and PN42,protein levels of CaMKⅡ in the entorhinal cortex in iodine-deficient,5ppm and 15ppm treatment groups were significantly lower than those of controls(P〈0.05).Conclusions Iodine deficiency and hypothyroidism may decrease the protein expression of CaMKⅡ in the entorhinal cortex of pups.
