摘  要:为了研究稻瘟病抗性基因Pi36的进化机理,以孟加拉国普通野生稻W39、马来西亚普通野生稻W40、印度野生稻L56为材料,对Pi36等位基因进行了克隆和序列分析。测序结果与已知的Pi36和测序品种日本晴以及"93-11"的基因序列进行比较。结果表明,编码区6个品种间的同源性为83%~99%,CC-NBS区域的同源性平均为91%;LRR区域为95%。并且还发现在野生稻W39和W40基因的第二个外显子区域有一个转座子插入。进化分析表明,6个品种从整体上分为3类,L56、日本晴和Pi36聚为一类,说明这3个品种的亲缘关系较近;W39和W40聚为另一类;"93-11"的基因聚为单独的一类,处于独立进化的模式。To study evolution mechanism of the blast resistance gene, Pi36 and its allele were cloned and sequenced from common wild rice W39, W40 and L56, which were originated from Bangladesh, Malaysia and India.The gene sequences were analyzed including known Pi36 and reference sequenced cultivars Nipponbare and 93-11.The results showed that the nucleotide identity of ORF was 83%~99% among the six cultivars, CC-NBS with 91% and LRR with 95%.Interestingly, a transposon element was inserted in the second exon of W39 and W40.Phylogenetic analysis showed that six genes were distributed into three groups, which were L56, Pi36 and Nipponbare as the first group, W39 and W40 as the second group, and 93-11 as the third group.
