摘 要:采用离乳Wistar大鼠21只,随机分为4组:对照组(5只)饲喂大鼠标准饲料,饮自来水;高氟组(5只)饲喂大鼠正常饲料,饮100mg/L氟化钠(NaF)去离子水;低碘组(5只)饲喂低碘饲料(定制),饮去离子水;高氟低碘组(6只)饲喂低碘饲料,饮100mg/L氟化钠(NaF)去离子水。在大鼠20月龄时,处死后采取肾脏,检测其DNA的损伤情况,研究高氟低碘对老年大鼠肾脏细胞DNA损伤的影响。结果表明,与对照组相比,高氟组、低碘组和高氟低碘组的老年大鼠肾细胞拖尾率比对照组显著增高(P<0.01);高氟组和低碘组老年大鼠肾细胞的拖尾长度比对照组增长,但统计学上无显著性差异(P>0.05)。高氟低碘组大鼠肾细胞的拖尾长度比对照组显著增长(P<0.01),并且比高氟组和低碘组的肾细胞拖尾长度明显增长(P<0.01)。高氟组的肾细胞拖尾长度比低碘组的长,但差异不显著(P>0.05);肾细胞DNA损伤分级结果显示,对照组肾细胞Ⅰ级损伤为64.29%,Ⅱ级损伤达到35.71%。高氟组和低碘组老年大鼠肾细胞DNA损伤主要是Ⅱ级损伤,分别为60.00%和40.74%。高氟低碘组的肾细胞DNA几乎全部损伤,并且主要是Ⅲ级损伤。所以,高氟、低碘、高氟低碘处理都加剧了大鼠肾脏细胞DNA的损伤。To study the effects of high-fluoride(high-F) and low-iodine (low-I) on DNA damage in the kindey of aged rats.21 weaned Wistar rats were randomly divided into 4 groups, including control group (feeding 5 rats) with standard diet and tap water, high-F group (feeding 5 rats) with normal diet and 100 mg/L NaF deionized water, low-I group(feeding 5 rats) with low-I diet and deionized water, high-F+ low-I group (feeding 6 rats) with low-I diet and 100 mg/L NaF deionized water.The samples of kindey were obtained by rats' decapitation at 20 months old and were made into unicellular liquid respectively for SCGE.Compared with the control group, the tailing rate of kindey cells in aged rats in groups high-F, low-I and high-F+low-I was significantly increased (P0.01) and the tail length of kindey cells in aged rats in groups high-F, low-I and high-F+low-I was longer.The classification results of DNA damage in kindey cells indicated that DNA damage in kindey cells in aged rats in group control were mainlyⅠ-class(64.29%) and Ⅱ-class (35.71%).DNA in kindey cells in group high-F and low-I was damaged, being mainly Ⅱ-class (60.00% and 40.74%).The cell damage in group high-F+low-I was almost damaged, being mainly Ⅲ-class.High-F, low-I, high-F+low-I promoted DNA damage in kindey cells in rats.
