摘 要:以国产基因枪JQ-700分别将GUS(β-葡糖醛酸苷酶)基因射入谷子和玉米的花粉,经人工授粉获得种子。在加卡那霉素(Kan)的培养液中筛选获得Kan抗性植株。对抗性植株进行GUS组织化学和Southern杂交检测,结果表明有外源基因的整合与表达。转基因谷子和玉米的频率分别为0.18%和0.05%~0.21%。Bombardment to both millet and maize pollen grains with plasmid pBI121 containing kanamycinresistant gene and GUS gene were performed. After bombardment , pollens of Jigu No.11 millet were pollinated onto the stigma of a male sterile line 91045. Seeds were harvested and germinated in a selective nutrient solution contaning kanamycin 50 mg/L. GUS assay and Southern hybridization of kanamycinresistant seedlings indicated the intergration and expression of foreign gene. The transformation frequency was 0.18%. In maize, the pollen grains of maize inbred line CA11 and Danyu No.13 were bombarded and used for pollination. R0 seeds were harvested and germinated in a nutrient solution containing Kanamycin 140 mg/L. GUS and Southern analysis confirmed the intergration and expression of the foreign gene. The transformation frequency was 0.05% ̄0.21%.
