摘  要:探讨大鼠脑微血管内皮细胞的体外培养方法,为研究猪链球菌引起脑膜炎的致病机理打下基础。采用脑组织匀浆、二次过筛及酶消化技术分离大鼠脑微血管段,对分离的脑微血管内皮细胞进行了体外培养、形态学观察、免疫荧光鉴定及生长曲线的MTT法测定。结果表明:倒置显微镜下,细胞具有单层"铺路石样"排列的典型特征,免疫荧光检测第Ⅷ因子相关抗原呈阳性,成功建立了大鼠脑微血管内皮细胞的体外培养方法。The methods of culturing rat brain microvascular endothelial cells (BMECs) were studied to build up the basis for the studies of pathogenesis of meningitis caused by Streptococcus suis. We used the homogenization,filtration an denzyme digestion technique to isolate microvessels from rat brain. The isolated endothelial cells were cutured, observed morphologically and identified by immunofluorescence. We also determined the growth curve of BMECs by MTF assay. The cells looked like"slabstone", a typical appearance of cultured endothelial cells under inverted microscope. Correlation antigens of VIB factor were positive by immunofluorescence. The results indicated that we have already succeeded in establishing the method of culturing rat brain microvascular endothelial cells(BMECs) in vitro.
