摘 要:利用正交实验优化pH敏感型PASP载药材料的制备工艺,选取酮洛芬为模型药物,通过测定药物释放度,考察了制备载药水凝胶的主次因素并确定了最佳工艺为PSI与己二胺物质的量比20∶1,NaOH溶液浓度1mol/L,NaOH与原料聚琥珀酰亚胺(PSI)物质的量比0.4,凝胶载药率15%。由Peppas方程可知,凝胶在模拟肠液中的释放规律是扩散和溶蚀综合作用的结果,而在模拟胃液中则为溶蚀控制的作用。Orthogonal experiment is used to optimize the technology of preparing the pH sensitive poly-aspartic acid(PASP) hydrogel. The results show that the optimized parameter of preparing the pH sensitive PASP hydrogel is as follows., the hydrolytic concentration of NaOH is lmol/L, the mol ratio of NaOH to raw material is 0. 4, the ratio of raw material to cross-linking agent is 20 : 1, and the ratio of drugloading of vector is 15%. The release behavior is fitted by the Peppas equation. The PASP hydrogel has an ideal pH sensitivity property. It facilitates the release of drug from hydrogel in the solution of high pH value, but hinders the release of drug from hydrogel in the solution of low pH value.
