人前脂肪细胞的体外培养

摘　　要：目的建立人前脂肪细胞培养及其在药物诱导下向脂肪细胞分化的细胞模型。方法取成人的腹部纯脂肪颗粒,采用消化法进行人前脂肪细胞的原代培养,并在培养液中加入胰岛素和地塞米松对细胞进行诱导。对培养细胞进行形态学研究,测定生长曲线,并用油红O染色法对前脂肪细胞向脂肪细胞的分化进行定性、定量研究。结果培养出的人前脂肪细胞呈梭形,成分均一,增殖旺盛。经胰岛素和地塞米松诱导后,通过油红O脂肪染色法测定,证明前脂肪细胞已向脂肪细胞分化。结论在成熟的脂肪组织中存在着可以增殖,并可以分化成脂肪细胞的前脂肪细胞,经诱导后可以分化成成熟的脂肪细胞。Objective To establish the cell culture of human preadipocytes in vitro and its application in drug - induced differentiation. Methods Pure adult human abdominal fat particles were digested by collagenase. The preadipocytes were collected centrifugally. The cells were mixed in the culture medium with insulin and dexamethasone. Cultured cells were morphologically studied to determine the growth curve and the determination of cell differentiation was carried out by oil red O staining, in which the fat drops deposited in cells were detected qualitatively and quantitatively with an enzyme - linked immunosorbent detector. Results The preadipocytes exhibited the spindle - like characteristics with a uniform composition and strong proliferation. Induced by insulin and dexamethasone, the cells were stained dark red by oil red O, suggesting that the fat particles were deposited in cells. Conclusion Preadipocytes exist in mature adipose tissue, which can be induced to differentiate into lipocytes in vitro.
