摘　　要：克隆猪GPR40(G protein-coupled receptor)基因,并对其生物信息学进行分析。结果表明,克隆的该基因(GenBank登陆号为EU366318)与已发表的人、大鼠和小鼠GPR40基因核苷酸序列同源性分别为85%、80%和82%,氨基酸序列同源性依次为86%、82%和83%,其编码的蛋白质分子量为22.59 ku,等电点(pI)9.54,包含215个氨基酸残基,有许多内切酶位点;结构分析发现,该基因无信号肽,但包含一个7次跨膜结构,其蛋白产物多集中在细胞膜上。表明该蛋白结合在细胞膜上作为许多信号分子受体,参与体内脂类代谢等。The porcine GPR40（G protein-coupled receptor） gene has been cloned,we analyzed the sequence of the gene and coding protein by the method of bioinformatics.The main results were summarized as follows： the determined sequence of the porcine GPR40 gene has been registered in the GenBank with the accession number of EU366318.Homologous alignment analysis showed that the porcine GPR40 gene has 85%,80% and 82% homology to that of human,rattus norvegicus and mouse,respectively;and the homology of protein to human,rat and mouse reached 86%,82% and 83%,respectively.Bioinformatics analysis showed that the molecular weight of GPR40 protein is about 22.59 ku,pI is 9.54 and the number of amino acid residue is 215 aa,and the gene contains many endonuclease sites.The gene does not contain a signal peptide,but it includes a seven transmembrane structure.The protein product of the gene mainly concentrated in the cell membrane.The protein of porcine GPR40 gene is a transmembrane protein containing seven transmembrane protein structures.As the receptor of many signal molecules,it combines in the cell membrane,and takes part in lipid metabolism.
