鲍曼不动杆菌协同凝集试剂的制备及初步应用

摘　　要：目的:以富含SPA的金黄色葡萄球菌与鲍曼不动杆菌的多抗血清结合,制备特异的协同凝集试剂,探索其在鲍曼不动杆菌快速检测中的初步应用。方法:鲍曼不动杆菌免疫日本大耳白兔制备多抗血清并经吸收处理。富含SPA的金黄色葡萄球菌(No 1800株)经甲醛灭活后,与该多抗血清结合,制备协同凝集试剂,用各种临床分离菌株尤其是其它种类的不动杆菌与自制的协同凝集试剂反应,对该试剂的特异性和准确性进行评价。对临床新分离菌株,以本法和生物梅里埃公司的API 20E生化鉴定板对比,并结合细菌药敏结果判定自制协同凝集试剂的快速诊断效果。结果:鲍曼不动杆菌免疫日本大耳白兔获得了高质量的多抗血清,特异性好,凝集效价达1:320;抗体致敏的金黄色葡萄球菌,作为协同凝集试剂,能与待测鲍曼不动杆菌产生明显的凝集现象,灵敏度较血清凝集试验高16倍,而对其它种类的不动杆菌以及其它种属的细菌均无凝集现象。与传统的生化鉴定相比,协同凝集试验可提前1～2天得到细菌鉴定结果,结果准确性良好。结论:以鲍曼不动杆菌的多抗血清致敏富含SPA的金黄色葡萄球菌作为协同凝集试剂,对鲍曼不动杆菌具有特异、灵敏、快速的检测效果,可以作为常规诊断方法的有益补充。Objective：To establish a coagglutination test for the rapid detection of Acinetobacter baumannii.Methods：The specific antiserum was prepared by rabbits immunized with Acinetobacter baumannii followed by absorption capture of nonspecific antibody.SPA-rich Staphylococcus aureus killed by formalin was combined with the specific IgG from the antiserum then used as coagglutination reagent.The rapid detection of Acinetobacter baumannii was performed with the coagglutination reagent and its advantages were compared with the traditional methods.Results：In the present study,the high quality antiserum with agglutination titer of 1：320 and high specificity of Acinetobacter baumannii was got from the immunized rabbits.The rapid detection method of Acinetobacter baumannii was established with the SPA-rich Staphylococcus aureus combined with the specific IgG used as coagglutination reagent.This coagglutination test has a sensitivity of 16 times higher than that of the serum agglutination and relatively satisfied specificity.Conclusion：With the SPA-rich Staphylococcus aureus combined with the specific IgG of the antiserum,the coagglutination test for the rapid detection of Acinetobacter baumannii was established,which provides a helpful supplement for the rapid diagnosis of this pathogenic bacteria.
