摘  要:目的构建一种细胞芯片,根据芯片表面固定的CD抗体与细胞膜表面CD抗原免疫结合原理捕获细胞,检测人正常胃黏膜细胞系GES-1、胃腺癌细胞系7901细胞表面CD抗原表达的差异。方法将64种CD抗体蛋白固定在化学修饰醛基玻片上制成抗体阵列,将GES-1、7901细胞用胶原酶消化制成单个细胞悬液,用吖啶橙荧光标记后与芯片进行孵育,洗去芯片上游离的细胞,激光扫描仪检测芯片上CD抗体各点捕获到细胞的荧光信号,以此判定GES-1、7901细胞膜表面CD表型情况。结果发现CD9、CD13、CD29、CD44、CD49e、CD55、CD71、CD95抗体点均捕获到GES-1、7901细胞;CD15、CD24抗体点仅捕获到GES-1细胞;CD77、CD133、CD103、CD64抗体点仅捕获到7901细胞。结论 CD15和CD24可能是人正常胃黏膜细胞系GES-1细胞的表面标志物,CD77、CD133、CD103、CD64可能是人胃癌细胞系7901细胞的标志物。Objective To construct a cell microarray based on specificity of antigen-antibody reaction and by fixing monoclonal antibodies against CD antigens(CD antibodies)on the microarray substrate,and to screen cell-surface CD antigens in normal gastric cell line GES-1 and gastric cancer cell line 7901.Methods A total of 64 kinds of CD antibodies were spotted on the aldehyde slide to construct the cell microarray.GES-1 and 7901 cells were isolated,labeled with fluorescence,and captured by the cell microarray.The cell microarray was scanned with scanner,and the signals were analyzed.Results GES-1 and 7901 cells were captured by CD9,CD13,CD29,CD44,CD49e,CD55,CD71,and CD95 antibodies.CD15 and CD24 antibodies captured only GES-1 cells,and CD77,CD133,CD103,and CD64 antibodies captured only 7901 cells.Conclusion CD15 and CD24 may be the surface marker of human normal gastric cells,and CD77,CD133,CD103,and CD64 may be surface marker of human gastric cancer cells.
