摘  要:目的通过研究通络救脑口服液对老年性痴呆(AD)模型大鼠学习记忆能力及突触素(SYP)表达的影响,探讨通络救脑口服液抗AD的可能机制。方法将SD雄性大鼠随机分为空白对照组、假手术组、模型组、盐酸多奈哌齐组以及通络救脑口服液低、中、高3个剂量(12、24、48mg/kg)组,采用海马立体定向注射凝聚态β-淀粉样蛋白1-40制备AD动物模型。药物干预4周后,采用Morris水迷宫测试大鼠寻找平台所需时间和通过原平台次数。应用免疫印迹法检测海马区SYP蛋白的表达,电镜观察海马组织超微结构的变化。结果与假手术组比较,模型组平均逃避潜伏期显著延长,跨越原平台位置次数明显减少(均为P<0.01);与模型组比较,通络救脑口服液3组平均逃避潜伏期均显著缩短,跨越原平台位置次数均明显增加(均为P<0.01)。与假手术组比较,模型组大鼠海马区SYP蛋白表达明显降低(P<0.05);与模型组比较,通络救脑口服液3组SYP蛋白表达均显著升高(P<0.05)。电镜观察结果显示:模型组大鼠海马神经元变性,通络救脑口服液能改善大鼠海马神经元病理改变。结论通络救脑口服液对AD大鼠的学习记忆功能减退具有改善作用,可能与其上调SYP蛋白的表达有关。Objective To investigate the possible mechanism of anti-Alzheimer's disease(AD) through studying the influences of Tongluojiunao Oral Solution(TOS) on the learning and memory ability and synaptophysin(SYP) expression in rat AD model.Methods Male SD rats were randomly divided into the blank group,sham-operation group,model group,donepezil hydrochloride group(DH group) and high-dose,mid-dose and low-dose TOS groups(12,24,48 mg/kg) .The rat model of AD was established by hippocampus stereotaxis injection of condensedβ-amyloid protein(β-AP 1-40 ) .After 4 weeks the period of find platform and times of passing through original platform were detected in rats by applying Morris water maze.The expression of SYP in hippocampus was detected by using Western blotting method,and the changes of hippocampus ultrastructure were observed with electron microscope.Results In the model group,compared with the sham-operation group,the average escape latency was longer significantly and times of crossing over original platform reduced significantly(all P0.01) .In all TOS groups,compared with the model group,the average escape latency was shorter significantly and times of passing through original platform increased significantly(all P0.01) .In the model group,comparedwith the sham-operation group,the SYP expression in hippocampus decreased significantly(P0.05) . In all TOS groups,compared with the model group,the SYP expression in hippocampus increased significantly(P0.05) .The outcomes of electron microscope showed neuronal degeneration in hippocampus in the model group,while TOS relieved the pathological changes of hippocampus neurons in rats.Conclusion TOS can improve the learning and memory ability in AD rats,which may be related to that it can up-regulate SYP expression.
