摘　　要：目的建立测定犬血浆中靛玉红质量浓度的反相高效液相色谱法。方法以0.1mol/L的醋酸铵(含0.5%冰醋酸)-甲醇(25:75)为流动相,非那雄胺为内标,血浆样品经正庚烷-乙醚的混合溶剂萃取后,采用反相高效液相色谱法测定。结果血浆中靛玉红质量浓度的线性范围为0.010～0.800μg/mL,最低检测限为5ng/mL,平均相对回收率为92.32%～105.81%,日内和日间精密度的RSD均小于11.40%。结论所建立的检测方法快速、准确、灵敏度高,可用于靛玉红的药代动力学研究。Objective To establish an RP-HPLC method to detect the concentration of indirubin in dog plasma.Methods The mobile phase consisted of 0.1 mol /L ammonium acetate solution（containing 0.5% acetic acid）-methanol（25：75,V /V） was used.Finasteride was used as the internal standard.The plasma samples were pretreated by mixed solvent of normal heptane and aether,then analyzed by RP-HPLC.Results The linear range of indirubin was 0.010-0.800 μg /mL.And the recovery rate was 92.32%-105.81%.Both the within-day and between-day RSD were less than 11.40%.The minimum detective limit was 5 ng /mL.Conclusion This established RP-HPLC method is rapid,accurate,stable and sensitive,which can be applied to the pharmacokinetic study of indirubin.
