摘 要:目的观察并对比分析四味归肝经虫类中药全蝎、蜈蚣、土鳖虫、水蛭水提物对人肝癌HepG2细胞株增殖抑制作用差别。方法采用液氮冻融法及丙酮沉淀法制备药物,根据回归方程计算半数抑制率(IC50),MTT法检测各药物对HepG2细胞的增殖抑制作用。结果全蝎、蜈蚣、水蛭水提物具有显著抑制HepG2细胞增殖作用,呈明显的量效关系,土鳖虫水提物作用不明显,各药物的IC50分别为全蝎7.02mg/ml,蜈蚣3.07mg/ml,水蛭9.83mg/ml,土鳖虫433.57mg/ml。结论全蝎、蜈蚣、水蛭取提物均能有效抑制人肝癌HepG2细胞增殖,其中蜈蚣作用最强,而土鳖虫提取物抑制作用不明显。Objective To analysis the inhibition effects of four Chinese herbs of insect with liver channel tropism,such as scorpion,centipede,ground beetle and leech etc,to HepG2 cell proliferation.Methods All aqueous extracts were prepared by methods of rapid freezen-thaw with liquid nitrogen,and following precipitation with acetone.The proliferation inhibition rates of four drugs were assayed by MTT method and the half inhibitive concentration(IC50)was calculated by regression equation.Results The significant effects of proliferation inhibition and dose-effect correlation,have detected in the scorpion,centipede and leech in four drugs,but no significant effects in ground beetle.The IC50 of scorpion,centipede,leech and ground beetle were 7.02mg/ml,3.07mg/ml,9.83mg/ml and 433.57mg/ml.Conclusions The Chinese herbs of insect of scorpion,centipede and leech had the significant effects of proliferation inhibition to HepG2 cells,centipede had the strongest effect,and ground beetle had no significant effect.
